摘 要:目的建立吴茱萸药材的HPLC指纹图谱,并同时测定其中去氢吴茱萸碱、吴茱萸碱、吴茱萸次碱和吴茱萸内酯的含量。方法采用phenomenex luna C_(18)色谱柱(250×4.6 mm,5μm),乙腈-0.02%磷酸水二元梯度流动相,检测波长225 nm。对10批吴茱萸药材进行了指纹图谱和含量测定研究。结果建立了吴茱萸的HPLC指纹图谱,标定了19个共有峰,并对其中4个成分进行了含量测定。去氢吴茱萸碱、吴茱萸碱、吴茱萸次碱和吴茱萸内酯分别在相应的浓度范围内线性关系良好,平均加样回收率分别为98.9%,98.9%,97.0%和98.8%。结论该方法所建立的吴茱萸指纹图谱特征性强、方法简便,结合4种主要成分的含量测定可更好的控制其质量,为吴茱萸药材的质量控制提供科学依据。Objective An HPLC method was developed to evaluate the quality of Evodia rutaecarpa through fingerprints and multi-components determination. Methods The analysis was performed on a phenomenex luna C_(18) column(250 × 4.6 mm, 5 μm), with mobile phase consisted of acetonitrile and 0.02% phosphoric acid in gradient mode. The detection wavelength was set at 225 nm. Collect 10 batches from different origins to assess the similarities according to fingerprint similarity evaluation system. Results The HPLC fingerprint was set up and 19 common peaks were identified. The linearity of dehydroevodiamine, evodiamine, rutaecarpine and limonin were all good. The average recovery was 98.9%, 98.9%, 97.0%, 98.8%, respectively. Conclusion The developed method provides a scientific basis for controlling the quality of Evodia rutaecarpa.
