摘 要:目的:观察养血清脑颗粒对血管性痴呆(VD)大鼠海马CA1区糖原合成酶激酶3β(GSK-3β)蛋白及β-连环蛋白(β-catenin)表达的影响。方法:72只实验SD大鼠随机分为假手术组、VD模型组(模型组)和养血清脑颗粒治疗组(治疗组),采用改良的Pulsinelli四血管阻断法制作VD大鼠模型,分别在术后1、2、4和8周采用免疫组化法和Western blotting法检测海马CA1区GSK-3β及β-catenin表达水平。结果:模型组各时间点GSK-3β蛋白表达增多,1周时即有多量表达,2周时大量表达,4周时达高峰,8周时开始下降但仍较高。与假手术组相比,模型组各时间点GSK-3β蛋白表达增多,有统计学意义(P<0.01)。各时间点β-catenin蛋白表达增多,但与假手术组相比,差异无统计学意义(P>0.05)。治疗组各时间点GSK-3β及β-catenin蛋白表达明显增多,与模型组相比,有统计学意义(P<0.01)。结论:养血清脑颗粒可通过上调GSK-3β及β-catenin蛋白的表达,发挥对VD的治疗作用。This study was aimed to observe the effect of Yang-Xue Qing-Nao(YXQN) granules on expressions of glycogen synthase kinase-3β(GSK-3β) and β-catenin in CA1 area of hippocampus among vascular dementia(VD) rats. A total of 72 SD rats were randomly divided into the sham operation group, VD group(model group) and the YXQN granules treatment group(treatment group). The VD rat model was prepared by modified Pulsineli's four-vessel occlusion. The expressions of GSK-3β and β-catenin in CA1 area of hippocampus were detected by immunohistochemistry and western blotting at the 1^st week, 2^nd week, 4^th weeks and 8^th week after VD model operation. The results showed that expressions of GSK-3βwere increased in the model group at different time points, which were many quantities of expression at the 1^st week, and a large number of expressions at the 2^nd week. It reached peak at the 4^th week; and began to decline but still higher at the 8^th week. Compared with the sham operation group, the expression of GSK-3β was significantly increased in the model group at different time points(P〈0.01). The expression of β-catenin was increased in the model group at different time points. However, there was no statistical significance compared with the sham operation group(P〈0.05). Compared with the model group, expressions of GSK-3β and β-catenin were significantly increased in the treatment group at different time points(P〈0.01). It was concluded that YXQN granules upregulated the expression of GSK-3β and β-catenin, which may be helpful to VD treatment.
