白藜芦醇对人肾细胞癌786-0细胞存活素及p27蛋白表达的影响

摘　　要：目的探讨白藜芦醇(Res)对人肾细胞癌细胞存活素、p27蛋白表达的影响。方法使用Res培养液(25μmol·L^(-1))作用人肾细胞癌786-0细胞12,24及48 h后,应用流式细胞术检测786-0细胞周期和凋亡情况,采用蛋白质免疫印迹法检测细胞存活素、p27蛋白的表达情况,以空白培养液作为对照组。结果实验组G_1期[(64.87±3.76)%]及G2期[(9.91±0.82)%]比例显著性降低(P<0.05),S期[(25.05±3.12)%]比例显著性升高(P<0.05);实验组在12,24和48 h等不同时间点的细胞凋亡率[(5.12±0.64)%,(5.71±0.49)%,(5.84±0.30)%]均明显高于对照组[(0.72±0.27)%,(0.93±0.25)%,(1.12±0.21)%,P<0.05];实验组存活素蛋白表达水平明显高于对照组(P<0.05),且随时间延长表达呈显著性升高(P<0.05);实验组p27蛋白表达水平明显低于对照组(P<0.05),且随时间延长表达水平呈显著性降低(P<0.05);实验组细胞存活素与p27蛋白表达之间呈负相关性关系(P<0.05)。结论 Res通过抑制p27蛋白的表达而上调存活素的表达水平,进而促进肾细胞癌786-0细胞的凋亡。Objective To investigate the effect of resveratrol（ Res） on the expression of survivin,p27 protein in human renal cell carcinoma 786-0 cells. Methods The cell cycle and apoptosis of 786-0 cells were detected by flow cytometry,and the expression of survivin and p27 protein of 786-0 cells were detected by Western blotting at time point of 12,24 and 48 h after human renal cell carcinoma 786-0 cells were cultured with Res（ 25 μmol·L^（-1）） and blank medium,respectively.Results The ratio in G_1 phase [（ 64. 87 ± 3. 76） %] and G_2 phase [（ 9. 91 ± 0. 82） %] in the experimental group decreased significantly（ P〈0.05）,and the ratio of S phase [（ 25. 05 ± 3. 12） %] increased significantly（ P〈0. 05）. The apoptosis rate of experimental group at 12,24 and 48 h [（ 5.12±0.64） %,（ 5.71±0.49） %,（ 5.84±0.30） %]was significantly higher than that of the control group [（ 0. 72 ± 0. 27） %,（ 0. 93 ± 0. 25） %,（ 1. 12 ± 0. 21） %,P〈0. 05]. The expression of survivin protein in experimental group was significantly higher than that of the control group（ P〈0. 05）,and the expression was significantly increased with time prolonging（ P〈0.05）.The expression of p27 protein in experimental group was significantly lower than that of the control group（ P〈0. 05）,and the expression level was significantly decreased in time-dependent manner（ P〈0. 05）. There was a negative correlation between the expression of survivin and p27 protein in the experimental group（ P〈0.05）. Conclusion Res could inhibit the expression of p27 protein and up-regulate survivin expression level,and promote the apoptosis of renal cell carcinoma 786-0 cells.
