摘  要:目的建立HPLC(高效液相色谱法)测定不同产地及不同部位茯苓中茯苓酸的含量。方法采用高效液相色谱法,Alltima-C18色谱柱(4.6 mm×250 mm,5μm);流动相乙腈-0.05%磷酸(78∶22);柱温20℃;流速1.0ml/min,检测波长210nm。结果在66.4~331.8μg/ml范围内峰面积具有良好的线性关系,回归方程Y=8.449X+1.4095(r=0.9995),平均回收率为100.8%,RSD(相对标准偏差relative standard deviation)值为1.3%。茯苓皮中茯苓酸的含量高于茯神,茯神中茯苓酸的含量高于茯苓。结论该方法简便,重复性,专属性好,为茯苓不同部位的质量控制与评价奠定了基础。Objective To establish an HPLC method for determination of content of pachymic acid in different medicinal parts of Poria cocos( Schw.) Wolf from different areas. Methods HPLC was used in the quantitative analysis by using Alltima C18 chromatography column and acetonitrile-0. 05% phosphoric acid( 78: 22) as a mobile phase. The flow rate of mobile phase was 1. 0ml / min,the detect wavelength was at 210 nm. Results The linear ranges for pachymic acid was 66. 4 ~ 331. 8μg / ml( r =0. 9994),the average recoveries was 100. 8%( RSD = 1. 3%). The contents of pachymic acid was highest in Poriae Cutis and lowest in Poria. Conclusion The method is proved to be simple,accurate and reliable,and can be used for quality control of different medicinal parts of Poria cocos( Schw.) Wolf.
