摘　　要：目的:通过考察不同工艺(原工艺、水提合煎、水提醇沉)所得宁神提取物对小鼠镇静、催眠作用及有效成分的影响,对院内制剂宁神方制备工艺方法进行优化。方法:用戊巴比妥钠阈上剂量睡眠实验和阈下剂量睡眠诱导实验,观察三种工艺宁神提取物的催眠作用,用地西泮、复方五味子糖浆进行平行对照实验。并对不同工艺得到的提取物的出膏率、特女贞苷、丹酚酸B和二苯乙烯苷含量进行测定。结果:三种工艺提取物的戊巴比妥钠阈下和阈上剂量实验的数据变化趋势一致,都反映了宁神提取物可延长小鼠的睡眠持续时间,增加小鼠的入睡数量。结论:证明了宁神方具有镇静、催眠作用。相比于原院内制剂工艺,水提合煎(NS2)提取物具有更优药效,为更优工艺。且为宁神方新药开发工艺优化提供了药理学实验依据。Objective： To compare the sedative and hypnotic effects and chemical components of Ningshen extract with different extraction technologies. Methods： The sleeping time and delitescence included by above threshold dose of pentobarbital sodium and sleeping number induced by below threshold dose of pentobarbital sodium were utilized to observe the hypnotic effect of different extracts in mice. Diazepam and extracts of Schisandra chinensis was applied as positive control drugs. Results： All the three extracts could obviously reduce spontaneous activity, increase sleeping time and sleeping number in mice, while thesleeping delitescence was shortened. Extracts of all the herbs decocted together （NS2） showed more effective. Conclusion： All the three extracts could induce sedative and hypnotic effects. The sleeping time and deliteseence included by above threshold dose of pentobarbital sodium and sleeping number induced by below threshold dose of pentobarbital sodium can be introduced to guide the screening of extraction process.
