摘  要:目的通过采用飞行时间质谱技术对插入缺失遗传多态性遗传标记的检验,探讨该技术对降解检材的DNA分型策略。方法提取切片DNA,在用STR试剂盒检测未能获得分型结果的情形下,采用多重PCR和飞行质谱技术对常染色体上的30个InDel位点进行分型检测。结果对于STR分型失败的切片,InDel分型获得成功。结论对于切片等微量、降解的生物学检材,若常染色体STR基因座分型失败,可尝试进行InDel位点的分型,以获得更多的遗传信息。Objective The method for DNA typing of insertion/deletion polymorphism markers from degraded DNA was explored based on the technology of time-of-flight mass spectrometry. Method Genomic DNA was prepared from slice samples, and multiplex PCR was carried out. When no typing results were obtained using STR kits, samples were further analyzed using 30 InDels by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF). Results Full profiles of InDels loci were obtained. Conclusion For typing of DNA from trace or degraded biological samples, analysis of InDel markers by MALDI-TOF can provide more genetic information even when STR genotyping fails.
