摘  要:目的利用基因工程小鼠研究ZNF436在压力超负荷诱导的心肌肥厚及心肌纤维化中的作用。方法将24只ZNF436基因敲除(ZNF436^(—/—),KO)小鼠和24只ZNF436野生型(wild type,WT)小鼠分别随机分为模型组和假手术组(n=12),通过主动脉缩窄术建立小鼠心肌肥厚模型,运用超声检测的方法评价手术4周后小鼠心功能,组织病理学方法评价心肌肥厚及纤维化程度,实时定量聚合酶链式反应在m RNA水平检测心肌肥厚标志物心钠肽、脑钠肽、肌球蛋白重链β及心肌纤维化标志物Ⅰ、Ⅲ型胶原纤维。结果主动脉缩窄术后4周,KO模型组与WT模型组及KO假手术组相比,KO模型组小鼠心室腔增大,短轴缩短率下降(P均<0.05,n=12),心肌肥厚和纤维化的病理学检测及有关标志物表达水平模型组高于假手术组(P均<0.05,n=12),其中KO模型组表达增加程度比WT模型组更为显著(P均<0.05,n=12)。结论 ZNF436基因可抑制心肌肥厚及纤维化,对心功能起保护作用。Objective To clarify the role of ZNF436 gene in cardiac hypertrophy and fibrosis induced by chronic pressure overload. Methods Twenty-four ZNF436 knockout (KO) mice and twenty-four wild-type ZNF436 (WT) mice were randomly divided into model group and sham group respectively. Cardiac hypertrophy model was established through aortic banding surgery( n= 12). Four weeks after the aortic banding operation, cardiac function of the mice were evaluated by ultrasonic diagnosis equipment, and the degree of cardiac hypertrophy and fibrosis were evaluated by histological detection. In addition, the markers of cardiac hypertrophy including atrial natriuretic peptide (ANP), brain natriuretic peptide (BNP) , β-myosin heavy chain (β-MHC ), and fibrosis markers ( Collagen Ⅰ , Collagen Ⅲ ) were detected by real-time quantitative polymerase chain reaction(PCR) at the mRNA level. Results Four weeks after the aortic banding operation, the physiological parameters and analysis of cardiac size and function of KO group were reduced significantly(P〈0.05 , n = 12), and the degree of cardiac hypertrophy and fibrosis were increased dramatically (P〈0.05 , n = 12) while compared with those of WT group.
