摘 要:目的:探讨七叶皂苷钠对大鼠急性肾热缺血再灌注损伤的影响及相关机制。方法:30只成年雄性Wistar大鼠,分为3组(n=10):假手术组(sham组),对照组(缺血再灌注组),治疗组(七叶皂甙钠组)在肾缺血45 min后完全恢复灌流。术前及术后24 h检测血清肌酐(Scr)、尿素氮(BUN)浓度;检测肾组织超氧化物歧化酶(SOD)活性,丙二醛(MDA)及髓过氧化物酶(MPO)含量;电镜下观察肾小管超微结构变化;光镜下观察肾组织病理学改变。结果:治疗组与对照组比较,血清Scr、BUN浓度有显著降低。治疗组与对照组比较,SOD活性升高,MDA浓度降低,MPO下降(P<0.05),超微结构及病理损伤明显减轻。结论:七叶皂苷钠能减轻肾热缺血再灌注损伤,其机制可能与减轻肾脂质过氧化反应和抑制细胞凋亡,减少炎症有关。Objective: To investigate the effects of sodium aescinat on the acute renal ischemia / reperfusion( I / R) injury in Rats. Methods: 30 adult male wistar rats were randomly divided into three groups with 10 animals in each group: group sham operation( S); group ischemia / reperfusion( I / R); treatment group( sodium aescinat). In the treatment group sodium aescinat was administered intravenously at reperfusion( 10 mg / kg). Blood samples were obtained for determination of blood creatinine( Scr) and urea nitrogen( BUN) concentrations before operation and at 24 hours after operation. The rats were killed at the 24 hours after operation and left kidneys were removed for determination of SOD activity and MDA and MPO concentrations. The changes of renal tissue were detected by light and electronic microscope. Results: Blood Scr and BUN concentrations of treatment group were significantly decreased by compared constantly with group I / R at the 24 hours after operation. MDA and MPO concentrations were decreased significantly,SOD activity was significantly increased in treatment group compared with group I / R at the 24 hours after operation( P 0. 05). Microscopic and electronic examination showed that the renal I / R injury was significantly attenuated by sodium aescinat. Conclusion:Sodium aescinat can attenuate renal damage induced by I / R by inhibiting oxidative stress.
