摘  要:[目的]通过观察胃祺饮对功能性消化不良(FD)大鼠胃肠动力,肠道菌群DNA表达的影响,来探讨胃祺饮治疗FD可能的作用机制。[方法]将72只SD大鼠随机分为正常组,模型组,莫沙必利组,胃祺饮低、中、高剂量组,每组各12只。采用复合病因造模法(夹尾刺激法+不规则喂养法)造模,连续14d,模型复制成功后连续给药14d,并进行胃排空、小肠推进实验,用Real-Time PCR方法测定粪便中乳酸杆菌、双歧杆菌DNA的表达量。[结果]1造模后大鼠胃内残留率升高,与正常组比较,差异具有统计学意义(P<0.01);小肠推进率下降,与正常组比较,差异具有统计学意义(P<0.05)。胃黏膜未见明显病理性改变,提示造模成功。2胃祺饮各剂量组及莫沙必利组胃内残留率均有所下降,小肠推进率均有所升高,与模型组比较,差异具有统计学意义(P<0.05);3胃祺饮可以增加粪便中乳酸杆菌、双歧杆菌DNA的表达(P<0.01)。[结论]胃祺饮具有改善复合病因造模法诱导的FD大鼠胃排空的作用,其机制可能通过调节胃肠动力,促进消化吸收功能,调整肠道内环境,从而调控了肠道微生态有关。[Objective]To explore the underlying therapeutic mechanisms of Weiqi Decoction(WD)on functional dyspepsia(FD),and to study the impact of WD on gastrointestinal motility and gut microbiota.[Methods]Seventy-two Sprague-Dawley rats were randomly divided into six groups(n=12for each group):Control group(C);model group(M);Mosapride fed group(MF);Low dose WD fed group(LWD);Moderate dose WD fed group(MWD);and,High dose WD fed group(HWD).FD was induced by constantly squeezing rat tails plus hunger or overeating for 14 days,and then 14 days of consecutive intervention was applied.Gastric emptying and intestinal transit function were detected;Fecal Lactobacillus and Bifidobacterium DNA levels were detected by Real-Time polymerase chain reaction(PCR)analysis.[Results]Less movement,gastric emptying(P〈0.01),and decreased intestinal transit function(P〈0.05)were detected in all FD models;no pathological changes of gastric mucosa were detected.After treatment,the rates of gastric emptying(P〈0.01)and intestinal transit function(P〈0.01)in all WD groups and MF group were increased;and similar rates of gastric emptying and intestinal transit function were found in HWD group and MF group.Fecal levels of Lactobacillus and Bifidobacterium were increased in all WD groups(P〈0.01).[Conclusion]WD can promote gastric emptying function in rats with Functional Dyspepsia by a compound method,whose mechanisms may be regulating gastrointestinal motility,promoting digestion and absorption function,adjusting intestines environment,then regulatily the gut microbiota.
