摘  要:目的探讨青蒿素(ART)对肿瘤坏死因子α(TNF-α)介导的细胞永生化表达下调(REIC)表达相关平滑肌增殖的影响。方法将大鼠随机分为假手术组、模型组、50 mg/kg ART组和100 mg/kg ART组。建立大鼠颈动脉球囊损伤模型,分离血管平滑肌细胞进行培养。Western blot检测REIC表达,荧光定量PCR检测引物。结果大鼠颈动脉球囊损伤后,REIC基因表达受到抑制,ART处理对大鼠动脉平滑肌细胞中REIC蛋白的表达影响极小。而TNF-α处理下细胞中REIC蛋白的表达水平受到显著抑制。ART对细胞进行预处理,能够显著减轻TNF-α导致的REIC蛋白表达抑制。而ART处理呈剂量依赖性上调REIC的表达。结论 REIC表达通过TNF-α介导参与平滑肌增殖过程,而且应用ART可以抑制平滑肌增殖过程。Objective To explore the influence of artemisinine (ART) on smooth muscle proliferation related to reduced expression of immortalized cells (REIC) induced by tumor necrosis factor α (TNF-α). Methods Rats were randomly divided into sham-operated group, model group, 50 mg/kg ART group, and 100 mg/kg ART group. Balloon-damaged rat carotid artery models were estabhshed. Vascular smooth muscle cells were isolated and cultured. REIC protein and mRNA expression were detected by Western blot and fluorescent quantitation PCR. Results The expression of REIC genes was suppressed when carotid arteries were balloon-damaged. Vascular smooth muscle ceils, intervened with ART, minimally affected REIC protein expression in rat arterial smooth muscle cells. While treated with TNF-α, the level of cellular expression of REIC protein was significantly restrained. Preconditioning of ART on cells would prominently ameliorate the TNF-α-induced suppression of REIC protein expression and unregulated REIC expression in a dose-dependent manner. Conclusion REIC is involved in the process of TNF-α-induced smooth muscle proliferation. ART is capable of suppressing proliferative process of smooth muscle cells.
