摘  要:目的:观察桃仁提取物对瘀热互结证大鼠抗微血栓效应,检测其与血小板黏附分子31(CD31),核转录因子-κB(NF-κB)及其抑制物κB(IκB)表达的关系,探讨桃仁对血瘀证抗血栓形成的机制。方法:60只SD大鼠随机分为正常组,模型组,桃仁提取物低、中、高剂量组(2,4,8 g·kg-1)及丹参组(14 g·kg-1),用角叉菜胶+干酵母复合注射法建立瘀热互结证模型,用药组分别按剂量ig给药,连续7 d。观察各组血瘀体征,全血黏度,血浆黏度及纤维蛋白含量;组织病理观察微血栓和肾损伤;免疫组化,原位杂交检测分析内皮细胞CD31,NF-κB,IκB蛋白及mRNA表达吸光度A。结果:与正常组比较,模型组大鼠出现血瘀体征:全身皮肤黏膜充血,肿胀,发绀;全血黏度,微血栓,肾损伤增多或明显;内皮CD31蛋白,NF-κB核蛋白,IκB总蛋白增多,NF-κB总蛋白显著减少(P<0.01)。与模型组比较,桃仁提取物高剂量组血瘀体征减轻,全血黏度,血浆黏度,微血栓及肾损伤均明显降低或减少(P<0.01),内皮CD31蛋白减少,NF-κB,IκB总蛋白及mRNA均明显增多(P<0.05,P<0.01)。结论:桃仁提取物能显著改善瘀热互结证,降低血黏度,抑制微血栓形成,保护肾实质;其可能通过降低CD31表达,促进NF-κB及其抑制物IκB的基因转录和翻译,或抑制其降解,从而发挥抗微血栓的作用。Objective: To study the antithrombotic effect of Persicae Semen extract( PSE) in blood stasis rats,its relationship with platelet adhesion molecule 31( CD31),nuclear transcription factor-κB( NF-κB)and its inhibitor κB( IκB) expressions and the antithrombotic mechanism of Semen Persicae against blood stasis syndrome. Method: Sixty SD rats were randomly divided into the normal group,the model group,the PSE low-,middle-,high-dose groups( 2,4,8 g·kg^(-1)) and Danshen group( 14 g·kg^(-1)). The stasis model was established through ip with carrageenan and dry yeast liquid. All of treatment groups were given drugs by oral administration.The stasis symptom,blood viscosity,plasma viscosity,fibrinogen,micro-thrombosis and renal damage were observed by blood viscometer and histopathological method. CD31,NF-κB,IκB protein and mRNA expression absorbance of vascular endothelial cells were detected by immunohistochemistry method and in situ hybridization technique. Result: Compared with the normal group,the model group had some stasis symptoms,such as congestion,swelling, cyanosis under skin mucosa; its blood viscosity, micro-thrombosis and renal injury increased,CD31,nuclear NF-κB and total-IκB protein of endothelial increased,but NF-κB total protein decreased( P〈0. 01). Compared with the model group,the PSE high dose group showed relieved stasis symptoms,lower blood viscosity,plasma viscosity,micro-thrombosis and relieved kidney damage( P〈0. 01),decreased CD31 protein of endothelial,but higher NF-κB,IκB total protein and mRNA( P〈0. 05,P〈0. 01). Conclusion: PSE can significantly improve blood stasis symptoms,lower blood viscosity,inhibit thrombosis and protect renal. It might show the anti-micro thrombosis effect by promoting NF-κB and its inhibitor IκB gene transcription and translation,or inhibiting its degradation,as well as reducing the CD31 protein.
