摘  要:为了制备含有咪唑基团的水凝胶,本文将反应制备的Im-PAA-HEAA大分子单体和聚乙二醇二丙烯酸酯(PEGDA)混合,在紫外照射下发生自由基反应,从而得到Im-PAA-HEAA/PEGDA水凝胶。在低浓度的锌离子溶液刺激下,水凝胶表现出体积收缩、吸水率下降和强度增强等响应性,并且表现出良好的细胞相容性。将尿刊酸接枝于聚乙烯亚胺(PEI),制成含咪唑的非病毒基因载体(Im-PEI),该载体可以有效复合pDNA,并具有低细胞毒性和较高的转染效率。利用锌离子和咪唑之间的络合作用,可以将Im-PEI/DNA复合物有效地固定于Im-PAA-HEAA/PEGDA水凝胶表面。研究结果表明:与无锌离子作用相比,Im-PAA-HEAA/PEGDA水凝胶在锌离子作用下能够吸附更多的Im-PEI/DNA复合物,该水凝胶有望在反向基因转染方面得到应用。In order to prepare hydrogel containing imidazole group,Im-PAA-HEAA macromer and polyethylene glycol diacrylate(PEGDA)were mixed homogeneously and then they were gelled to yield Im-PAA-HEAA/PEGDA hydrogel through radical reaction in UV radiation.Hydrogels show obvious volume shrinkage,water absorption reduction and mechanical enhancement in low concentration of zinc ion solution and exhibit excellent biocompatibility.A non-viral gene vectors Im-PEI which can combine DNA effectively and shows low cytotoxicity and high gene transfect efficiency is synthesized by grafting urocanic acid onto PEI.Due to the complexing force between the Zn-(2+) and imidazole,the Im-PEI/pDNA can be adsorbed on the surface of hydrogels with imidazole.The results indicate that Im-PAA-HEAA/PEGDA hydrogels can absorb more Im-PEI/DNA complexes by zinc ions complexion than these without zinc ions.Hydrogels may have great potential in reverse gene transfection.
