摘 要:为探讨贵州省草海鲫鱼和普安鲫鱼群体的遗传变异和系统进化,对其rDNA ITS-1序列进行PCR扩增、测序分析。结果表明:24尾草海鲫ITS-1序列长度具有明显的多态性,分为296-297bp、300-301bp和337-339bp 3种类型;共发现1个多态位点、3个碱基插入和1个碱基缺失,其GC含量(70.8%)明显高于AT含量(29.2%)。14尾普安鲫ITS-1序列长度仅有1种为337-339bp,共检测到18个多态位点,存在1个碱基缺失和1个碱基插入,其GC含量为71.2%,远远高于AT含量(28.8%)。所有序列共产生11种单倍型,单倍型的平均遗传距离(P)以及平均核苷酸差异系数(K)草海鲫低于普安鲫;Tajima’s D和Fu and Li’s D中性检验表明,草海鲫未经历种群扩张,而普安鲫可能经历种群扩张。普安鲫遗传多样性较草海鲫丰富。In order to discuss the genetic mutations and phyletic evolution of two populations,C.auratus in Caohai Lake and in Pu'an,PCR amplification and sequencing analysis were conducted on their rDNA ITS-1sequence.Results:24individuals of Caohai C.auratus had three obviously different length types of ITS-1that could be divided as 296-297bp,300-301bp and 337-339bp.Meanwhile,1polymorphic sites,3bases insertion and 1bases deletion was found.The base of GC content(70.8%)was visibly higher than that of AT content(29.9%).14 individuals of Pu'an C.auratus had only one sequence type,337-339bp,which had been detected out 18 polymorphic sites,1insertion base and 1deletion base.The GC content(71.2%)was far more higher than that of AT content(28.8%).Eleven kinds of new haploid types were produced by the pooled samples.The average genetic distance of haploid types and nucleotide diversity index of C.auratus in Caohai Lake were lower than that of C.auratus in Pu'an.Tajima's D,Fu and Li's D indicated that the Caohai C.auratus had not been undergone a population expansion,but they indicated a possible population expansion in Pu'an C.auratus.C.auratus in Pu'an has a quite more abundant genetic diversity than C.auratus in Caohai.
