摘  要:为了筛选红托竹荪最适培养基与培养条件,提高红托竹荪菌种的生产效率,利用单因素试验及正交试验分别对红托竹荪培养基及培养条件进行优化。结果表明:葡萄糖为最适速效碳源,果糖为最适迟效碳源;蛋白胨为最适速效氮源,硫酸铵为最适缓释氮源;最佳C/N比为23;ZnSO4、竹叶煮汁、VB6和IAA能显著地促进菌丝的萌动与生长;最适pH为6.0,最佳培养温度25℃且避光,但28℃能刺激菌丝快速萌动。结论:影响菌丝生长的因素主次关系为IAA>竹叶>速效与迟效氮源的配比>VB6>速效与迟效碳源的配比,最优组合葡萄糖5g,果糖25g,蛋白胨2.5g,硫酸铵2.5g,竹叶20g(煮汁),VB60.16g,生长素(IAA)1.6mg,水1000mL。The medium and cultural conditions of D.rubrovolvata were optimized by single-factor test and orthogonal test to screen the optimization medium and cultural conditions for improving production efficiency of D.rubrovolvata strains.Results:Glucose and fructose are the optimum rapid-acting and slow-acting carbon source but peptone and ammonium sulfate are the optimum rapid-acting and slowrelease nitrogen source respectively.The optimum C/N ratio is 23.ZnSO4,bamboo leaf extract,VB6 and IAA can improve hyphal germination and growth significantly.The optimum pH and cultural temperature are 6.0and 25℃ under the dark condition separately but 28℃can stimulate rapid germination of mycelia.In conclusion,factors affecting mycelia growth are IAA〉 bamboo leaf extract〉rapid-acting/slow-acting nitrogen source ratio 〉VB6〉rapid-acting/slow-acting carbon source ratio.The optimum medium components include 5g glucose,25 g fructose,2.5g peptone,2.5g ammonium sulfate,20 g bamboo leaf extract,0.16 g VB6,1.6mg IAA and 1 000 mL water.
