摘 要:目的观察反复短暂肢体缺血对高脂饮食兔血液系统及造血功能的影响。方法新西兰白兔28只随机分为正常组、高脂组、假缺血组和缺血组,每组7只。正常组饲以正常饲料,其余3组采用高脂饲料,其中高脂组不做任何处理,假缺血组除不造成缺血外其他处理同缺血组,缺血组在高脂饮食的同时给予每天一次的反复短暂肢体缺血:用血压计袖带环扎兔左大腿根部加压至200mmHg造成肢体缺血5min、松开袖带使肢体再灌注5min、重复6个循环。实验12周后取外周血检测血红蛋白、红系细胞数、白系细胞数和血小板数,取骨髓涂片进行细胞分类计数分析。结果缺血组、高脂组和假缺血组与正常组相比,外周血血红蛋白、红细胞计数均出现一定程度的下降,网织红细胞比例出现一定程度的上升(P〈0.05);但前3组相比差异无统计学意义(P〉0.05)。缺血组与其他3组相比,骨髓中的中幼红细胞百分比增加(P〈0.05),中性粒细胞、中性中幼粒细胞、嗜酸性粒细胞、嗜酸性中幼粒细胞以及嗜酸性晚幼粒细胞的百分比均减少(P〈0.05),粒红比值明显下降(P〈0.05)。结论持续12周的反复短暂肢体缺血对高脂饮食兔外周血不产生明显的影响;可能会在一定程度上促进高脂饮食兔骨髓红系增生、抑制中性粒系和嗜酸性粒系的增生。Objective To investigate the effects of regular transient limb ischemia on hematological system and hematopoietic function in rabbits fed with high-fat diet. Methods Twenty-eight New Zealand White rabbits were randomly divided into four groups: control group, high-fat diet group, sham ischemia group and ischemia group, each group had seven rabbits. Control group were fed with normal diet, the other three groups were fed with high-fat diet. No other procedure was performed to the rabbits in high-fat diet group. Six cycles of regular transient limb ischemia induced by inflating a blood pressure cuff placed on the left lower limb to 200 mmHg for 5 min followed by deflating the cuff for 5 min, was performed once per day in the rabbits of isehemia group during the 12-week period, while the rabbits in sham ischemia group had a deflated cuff accordingly. After 12 weeks, peripheral blood samples from all the rabbits were analyzed for hemoglobin measurement, red blood cell count, white blood cell count and platelet count, bone marrow smears from il- iac bone were analyzed for bone marrow cell analysis. Results Compared to the control group, peripheral blood hemoglobin and red blood cells in ischemia group were decreased, the percentage of reticuloeytes was increased (P〈0.05). However, these variables have no significant differences among high-fat group, sham ischemia group and ischemia group (P〉 0.05). Compared with the other three groups, the percentage of polychromatic erythroblast in the bone marrow in ischemia group was increased (P〈0.05), the percentages of neutrophils, neutral myeloeytes, eosinophils, cosinophilic myelocytes and eosinophilc metamyelocytes in the bone marrow were signifi- cantly reduced in ischemia group (P〈0.05), and the myeloid/erythroid ratio in ischemia group was significantly decreased (P〈0.05). Conclusion Twelve weeks of regular transient limb isehemia has no significant effect on the peripheral blood in rabbits fed with highfat diet, but it may promote erythroid
