丹参片和丹参注射液对多柔比星在大鼠体内药动学的影响

摘　　要：目的:研究联用丹参片/丹参注射液对多柔比星在大鼠体内药动学的影响。方法:18只SD大鼠随机均分成A、B、C 3组。A组大鼠左颈iv多柔比星8 mg/kg;B组:左颈iv丹参注射液0.5 ml,0.5 h后左颈iv多柔比星8 mg/kg;C组大鼠ig丹参片混悬液1ml(18片溶于50 ml纯水),7 d后左颈iv多柔比星8 mg/kg。3组大鼠分别于给药后0.083、0.25、0.5、0.75、1、2、4、6、8、12、24 h于右颈静脉取血0.6 ml制备血浆,高效液相色谱法测定其血药浓度,3p97软件计算药动学参数,比较3组药动学参数的差异。结果:与A组比较,B组CL、Vss升高,AUC0-t降低(P<0.05);C组Vss降低(P<0.05),k10增加、t1/2β减小。结论:丹参片/丹参注射液均可提高多柔比星进入各组织的分布速度,两药联用可能会提高肿瘤部位的药物浓度。OBJECTIVE：To study the effects of Danshen tablets or Danshen injection combination on the pharmacokinetics of doxorubicin in rats in vivo. METHODS：18 SD rats were randomly divided into group A,B and C. Group A was given doxorubicin8 mg/kg intravenously via left jugular trunk;group B was given Danshen injection 0.5 ml intravenously via left jugular trunk,and0.5 h later given doxorubicin 8 mg/kg intravenously via left jugular trunk;group C was given Danshen suspension 1 ml（18 tablets dissolved in 50 ml pure water）intragastrically,and 7 d later given doxorubicin 8 mg/kg intravenously via left jugular trunk. 0.6 ml venous blood were extracted from the right jugular vein 0,0.083,0.25,0.5,0.75,1,2,4,6,8,12 and 24 hours after medication. The plasma concentration of doxorubicin was determined by HPLC;the pharmacokinetic parameters were calculated by using3p97 software;the difference of pharmacokinetic parameters were compared among 3 groups. RESULTS：Compared with group A,CL and Vssof group B increased while AUC0-tdecreased（P〈0.05）;Vssof group C decreased（P〈0.05）,k10 increased and t1/2βdecreased. CONCLUSIONS：Danshen tablet/Danshen injection can increase the distribution speed of doxorubicin into tissues,and both can improve the concentration of drug in tumor focus.
