缬沙坦对心梗大鼠心脏血管紧张素转换酶2表达的影响

摘　　要：目的研究缬沙坦对大鼠心梗后心脏血管紧张素转换酶2(ACE2)表达的影响,以探讨心梗后心脏重构及其治疗的新机制。方法将冠脉结扎术后24h的SD大鼠随机分为心梗组、小剂量缬沙坦组(10mg·kg^(-1)·d^(-1))和大剂量缬沙坦组(30mg·kg^(-1)·d^(-1)),另设假手术组。灌胃给药4周后测定左室非梗死区血管紧张素Ⅱ(AngⅡ)水平、左室非梗死区ACE2mRNA水平。结果与假手术组相比,心梗组心脏非梗死区AngⅡ、ACE2mRNA水平显著增高(P<0.05)。两种剂量缬沙坦都可明显降低心梗组的AngⅡ水平(P<0.05),增高左室非梗死区ACE2mRNA水平(P<0.05),且大剂量缬沙坦的上述作用较小剂量更显著(P<0.05)。结论缬沙坦能增强心梗大鼠心脏ACE2的表达,且随剂量的增大而增强,为研究心梗后心室重构的防治提供了新思路。Objective To investigate the changes of ACE2 expression in the noninfarcted area of the ventricle after acute myocardial infarction （AMⅡ） in rats and also the effects of valsartan on ACE2 expression. Methods Twenty-four hours after the operations of ligation on the left anterior descending branch （LAD） of coronary artery, the surviving SD rats were randomly divided into three groups： AMI group,, low-dose valsartan treatment group （10 mg · kg^-1 · d^-1） and high-dose valsartan treatment group （30 mg· kg^-1 · d^-1）. The sham-operated group was also established. After four weeks of treatment, the following items were measured ： Ang Ⅱ levels of noninfarcted area of the ventricle ; ACE2 mRNA levels in the noninfarcted area of the ventricle. Results Compared with the sham-operated group, cardiac AngⅡ , ACE2 mRNA levels of AMI group increased signifi cantly （P〈0.05）. Both high and low dose valsartan decreased cardiac Ang Ⅱ of AMI group significantly （P〈0.05） and increased ACE2 mRNA levels significantly （P〈0.05） while the high-dose valsartan had a more significant effect than the lowdose （P〈0.05）. Conclusion Valsartan could increase cardiact ACE2 expression in AMI rats and the high-dose valsartan had a more significant effect than the low-dose.
