摘 要:以不结球白菜(Brassica campestris ssp.chinensis Makino)"矮萁苏州青"子叶为外植体,研究了激素浓度和AgNO_3浓度组合对不结球白菜离体培养的影响.结果表明:诱导不定芽形成的最佳培养基为MS+0.3 mg/L NAA+5 mg/L 6-BA;在此基础上添加5 mg/L AgNO_3可显著提高不定芽的诱导频率,芽诱导频率高达31.3%;生根最佳培养基为含有0.3 mg/L NAA的1/2MS培养基.该研究初步建立了"矮萁苏州青"的植株再生体系,为用基因工程手段开展不结球白菜分子育种奠定了基础.The cotyledon of non-heading Chinese cabbage( Brassica campestris ssp. chinensis Makino) was used as explants to investigate the effects of different concentrations of hormone and of AgNO_3 on its culture in vitro based on one variety ″short broom suzhouqing″. The results showed that the induction rate of adventitious bud was the highest on the MS medium containing0. 3 mg / L NAA and 5 mg / L 6-BA,while 5 mg / L AgNO_3 added on this medium could efficiently improve the regeneration rate( high to 31. 3%). Meanwhile,half strength MS supplemented with 0. 3 mg/L NAA was the best medium for rooting of non-heading Chinese cabbage. Our study set up a primary regeneration system for non-heading Chinese cabbage short broom suzhouqing.This system laid the foundation of molecular breeding of non-heading Chinese cabbage using genetic engineering further.
