摘　　要：高尔基磷酸化蛋白2(Golgi phosphoprotein 2,GOLPH2)是一种与多种疾病密切相关的高尔基体膜蛋白,研究该蛋白的结构、功能、在正常组织和肿瘤组织中的表达调控机制及其与疾病的关系等具有十分重要的应用价值。文章采用转录激活样效应因子核酸酶(transcription activator-like effector nuclease,TALEN)技术完成GOLPH2基因敲除小鼠模型的构建及其基因型分析。通过TALEN识别位点确定、TALEN质粒对的设计与构建、TALEN体外转录、TALEN mRNA受精卵注射及胚胎移植等技术手段和方法,获得编码GOLPH2序列出现移码的纯合子雄鼠(F0)。通过若干代繁殖稳定获得GOLPH2敲除纯合子小鼠。Golgi phosphoprotein 2 （GOLPH2） is a type II Golgi transmembrane protein related with many diseases. The studies on the structure, function, mechanisms of its regulation in normal and neoplastic tissues, and its relation with diseases are of significance, both in science and applicatiorL In this paper, the generation of GOLPH2 knock-out mouse by transcription activator-like effector nuclease（TALEN） technology and geno- type analysis of it are reported. Male mice（F0 ） with frarneshih of GOLPH2 are obtained by knock-out site determination, construction of TALEN plasmid, TALEN transcription in vitro, TALEN mRNA fertilized egg injection and embryo transplantation. Through continuous reproduction, homozygous mice with GOLPH2 knock-out are steadily obtained.
