摘  要:为研究重组鸡白细胞介素-6-2融合蛋白(rChIL-6-Linker-ChIL-2)对鸡体免疫功能的影响,采用MTS法检测重组融合蛋白在28日龄SPF鸡体内接种后不同时间对外周血淋巴细胞(PBLC)中T淋巴细胞和脾B淋巴细胞增殖活性的影响,采用流式细胞术检测它对鸡PBLC中CD3+CD4+T淋巴细胞、CD3+CD8+T淋巴细胞百分含量及CD4+/CD8+比值的影响。结果显示,接种后第7~35天,重组融合蛋白可显著提高鸡体PBLC中T淋巴细胞和脾中B淋巴细胞的增殖活性,两者活性分别高于单一rChIL-2蛋白或单一rChIL-6蛋白对照、低于或接近于rChIL-6蛋白+rChIL-2蛋白混合物。接种后第7~21天,重组融合蛋白可显著提高CD3+CD4+T淋巴细胞百分含量、降低CD3+CD8+T淋巴细胞百分含量,显著提高CD4+/CD8+比值,其活性显著高于单一rChIL-6蛋白或单一rChIL-2蛋白对照,但与rChIL-6蛋白+rChIL-2蛋白混合组差异不显著。结果表明,重组融合蛋白在鸡体内可显著促进T、B淋巴细胞增殖,提高CD3+CD4+T淋巴细胞百分含量和CD4+/CD8+比值,从而增强鸡体的体液免疫和细胞免疫功能;重组融合蛋白在体内发挥了ChIL-6和ChIL-2的协同作用活性。In order to explore the effect of a recombinant chicken interleukin-6-2fusion protein(rChIL-6-Linker-ChIL-2protein)on the immune function of chicken,the MTS method was used to detect the proliferation activity of T and B lymphocytes in 28days-old SPF chickens,and flow cytometry was used to detect the percentage of CD3+CD4+T-lymphocytes subsets,CD3+CD8+T-lymphocytes subsets and the CD4+/CD8+ratio in PBLC on different days post-injection(dpi)in the 28-day-old SPF chicken,respectively.The results showed that,from 7dpi to 35 dpi,the recombinant fusion protein could significantly improve the proliferation activities of T-lymphocytes in PBLC and B-lymphocytes in the spleen,and both of which was higher than the single rChIL-2or single rChIL-6protein control,lower than or close to rChIL-6 +rChIL-2protein mixture.From 7dpi to 21 dpi,the recombinant fusion protein could significantly improve the percentage of CD3+CD4+T-lymphocytes subsets,reduce the percentage of CD3+CD8+T-lymphocytes subsets,and significantly increase the CD4+/CD8+T-lymphocytes ratio.The activities of the recombinant fusion protein were significantly higher than that of either single rChIL-6protein or single rChIL-2protein control,but showed no significant difference with that of the rChIL-6+rChIL-2 mixture protein control group.These results indicated that the recombinant fusion protein could significantly promote the proliferation activities of T and B lymphocytes,increase the percentage of CD3+CD4+T-lymphocytes subsets and CD4+/CD8+ratio,thereby enhance humoral and cellular immune function,which suggest that recombinant fusion protein plays a synergistic role of interleukin-6and interleukin-2activity in vivo.
