藏药二十五味珊瑚丸中马兜铃酸A及汞盐等重金属含量测定

摘　　要：藏药二十五味珊瑚丸处方中的木香在流通和使用过程中存在与含马兜铃酸的青木香混用现象。文章建立RP-HPLC法（流动相：甲醇-0.05%冰醋酸（62：38）;检测波长：250nm;柱温：30℃;流速：1m L/min）测定处方中马兜铃酸A的含量,并采用2010年版《中国药典》重金属检查法第二法、硫氰酸铵滴定法,分别测定了二十五味珊瑚丸处方样品中的重金属总量和硫化汞的含量,采用一般鉴别试验对可溶性汞盐进行检查,控制其质量,保证临床使用安全性。马兜铃酸A和可溶性汞盐在样品中均未检出;所检测样品中重金属总含量在200~225ppm之间;硫化汞平均含量为29.75 mg·g-1,RSD为0.56%。结果表明,藏药二十五味珊瑚丸中木香的来源控制较好;重金属总含量较高,有必要进一步分析重金属成分是否为本品的活性必需成分以及是否具有毒性。本研究中硫氰酸铵滴定法测定硫化汞的含量方法简便、结果准确、重复性好,可用于藏药二十五味珊瑚丸的质量控制。The phenomenon of mixing useing between Saussurea lappa in the Ershiwuwei prescription and radix aristolochiae in aristolochic acid existed in their utilization and circulation. The article established a method of RP-HPLC（Mobile phase： Methanol-0.05% Glacial acetic acid 62-38%; detection wavelength： 250nm; Column temperature： 30℃; flow rate： was 1.0ml/min） and detected the contend of Aristolochic Acids A in prescription,and the second method for inspection of heavy metal from ＂Chinese Pharmacopoeia＂ 2010 edition and titration of Ammonium thiocyanate were used to detect the contents of total heavy metals and mercury sulfide in Ershiwuwei Shanhuwan. The soluble mercury salts was inspected with general identification test and controlled its quality to ensure the safety of clinical application. Aristolochic acid A and soluble mercury salts were not detected in the sample, but the total content of heavy metals was between 200 ~ 225 ppm, the average content of mercury sulfide was 29.75 mg/g and RSD was 0.56%. The result showed that the source of Saussurea lappa controlled well in the Ershiwuwei Shanhuwan but the total content of heavy metals was high. Therefore, it is necessary further analysis to detect whether it is the active ingredient of the product or toxic substances. In this study, ammonium thiocyanate titration methed used for testing the content of mercuric sulfide is very simple and reproducible with accurate result,and it can be used for controling quality of Ershiwuwei Shanhuwan.
