摘  要:为研究草莓中几丁质酶基因的转录特性及其在果实中对生物及非生物胁迫的响应,以"阿尔比"草莓为试验材料,运用实时荧光定量PCR技术对4个几丁质酶基因FaChi1-FaChi4在果实发育过程及不同器官中的表达情况及其对生物及非生物胁迫的响应情况进行研究。结果表明:1)FaChi4主要在果实中表达,在转色期和红熟期有2个表达峰,它在果实发育与成熟过程中的表达水平显著地高于FaChi1-FaChi3。此外,FaChi1主要在营养器官中表达,其中在叶中的表达量最高;FaChi2主要在叶中表达;FaChi3在各个器官中的表达量均相对较低。2)干旱胁迫及灰霉菌处理后,果实中FaChi1-FaChi4的表达均被诱导。外施脱落酸处理可诱导FaChi2与FaChi4的表达。以上结果说明:FaChi1-FaChi4在草莓果实发育过程中有不同的表达模式且具有器官特异性,其中FaChi4是在果实中表达的主要基因;FaChi1-FaChi4在果实对生物及非生物胁迫的响应中有一定的调节作用。To investigate the transcription characterization of chitinase in strawberry and their responses to biotic and abiotic stress factors in fruit,expression pattern of FaChi1-FaChi4 during fruit development and in various organs and their responses to biotic and abiotic stress factors in fruit of strawberry‘Albion'were investigated by RT-PCR.Results showed that:1)FaChi4 expressed mainly in the fruit,with two peaks at turning stage and red ripening stage and its expression level was apparently higher than that of FaChi1-FaChi3.In addition,FaChi1 expressed mainly in nutritive organs and was highest in the leaf;FaChi2 expressed mainly in the leaf;FaChi3 expressed at a relative lower level in each organ.2)FaChi1-FaChi4 in the fruit was up-regulated under drought stress and inoculation with Botrytis cinerea.FaChi2 and FaChi4 were induced after treatment with exogenous abscisic acid.The above results indicated that transcription of FaChi1-FaChi4 had different patterns during fruit development and was organ-specific,among which,FaChi4 was the main gene during fruit development;FaChi1-FaChi4 played a certain regulation role in the responses to biotic and abiotic stresses in fruit.
