摘 要:目的探讨依达拉奉联合血必净对兔肠梗阻手术后肺损伤的影响及机制。方法 36只成年健康大白兔随机分为正常对照组(C组)、模型组(M组)和依达拉奉联合血必净组(EX组),每组12只,制作可复性机械性完全性肠梗阻模型,梗阻24h后麻醉行回盲部手术切除术,术前EX组给予依达拉奉联合血必净静滴,C组和M组给予等量生理盐水。术后24h处死家兔,检测其血清和肺组织匀浆中丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性、谷胱甘肽过氧化物酶(GSH-PX)活性,血清中TNF-α、IL-1和热休克蛋白70(HPS70)水平,并计算肺干/湿重比值(D/W)。结果 EX组血清及肺组织中MDA含量低于M组,SOD和GSH-PX活性高于M组(P<0.05);EX组血清TNF-α和IL-1水平均低于M组,HPS70水平高于M组(P<0.05);EX组肺D/W高于M组(P<0.05)。结论依达拉奉与血必净通过抑制炎症反应保护家兔肠梗阻术后损伤的肺组织。Objective To investigate the effects of edaravone and Xuebijing on the lung injury after intestinal obstruction surgery in rabbits and to explore the probably mechanism. Methods Thirty- six healthy adult rabbits were randomly divided into three groups: normal control group( group C),model group( group M) and edaravone combined with Xuebijing group( group EX),each group with 12 rabbits. Hone mechanical model of thecompleteintestinalobstructionwereestablishedwithreferencetothemethodofLilinect,)Resectionoftheileocecaljunctionoftherabbitwasperformedafter24hours.Edaravonecombining with Xuebijing were given in group EX,while normal saline were given in group C and group M. After operation 24 h rabbits were executed,dry / wet weight( D / W) ratio of lung was counted and malondialdehyde( MDA) content,superoxide( SOD) activity,glutathione peroxidase( GSH- PX) activity in serum and lung were detected,respectively. In addition,the levels of tumor necrosis factor- α( TNF- α),interleukin- 1( IL-1),hea proteint shock 70( HPS70) of serum were determined. Results The MDA content of serum and lung and the level of TNF- α,IL- 1 in group M were significantly higher than those in group C and group EX( P〈0. 05). However,D/W ratio of lung,SOD activity,GSH- PX activity of serum and lung,HPS70 of serum in group M were significantly lower than those in group C and group EX( P〈0. 05). Conclusion Edaravone combined with Xuebijing can alleviate lung injury after intestinal obstruction surgery by inhibiting inflammatory mediators to relieve lung damage from oxygen free radicals which are generated by intestinal ischemia reperfusion.
