摘 要:目的乙醇梯度沉淀白芨多糖,以期获得不同分子量段的多糖,并对梯度分离效果进行了初步评价。方法水提醇沉法制备白芨粗多糖,采用不同浓度乙醇对所得白芨粗多糖进行梯度沉淀,测定并计算各级产物的黏均分子量和总多糖含量。结果水提醇沉所得白芨粗多糖中总糖含量为67.62%,收率为34.70%。梯度醇沉后所得各分级产物的黏均分子量随乙醇浓度升高而降低。紫外和红外图谱均显示各分级产物与粗多糖波形近似。结论梯度醇沉法用于白芨多糖的分子量分级简便易行、高效可控,并兼有纯化效果。Objective To fractionate and evaluate polysaccharides with different molecular weight( MW)from crude Bletilla striata polysaccharide( BSP) by ethanol precipitation method. Methods Crude BSP was extracted by water extraction and alcohol precipitation method,and then fractioned by ethanol precipitation method. The obtained BSP fractions were then analyzed on relative MW,content of total polysaccharides,and characterized by UV and IR spectrometry as well. Results The content of total polysaccharides of crude polysaccharide from Bletilla striata was 67. 62%,and the yield was 34. 70%. As ethanol concentration increasing,the average MW of obtained BSP fraction was progressively decreased. Both UV and IR spectrum indicated that there was almost no difference on chemical structure between crude BSP and BSP fractions,but the BSP fractions with higher purity. Conclusion The ethanol precipitation method provides a simple but rapid technique for purification and fractionation of polysaccharide from Bletilla striata.
