摘  要:目的观察桂枝汤对糖尿病大鼠心脏自主神经病变重塑的影响及可能作用机制。方法 24只大鼠随机分为正常对照组、模型对照组、弥可保组和桂枝汤组,每组8只。除正常对照组外,其余大鼠采用一次性腹腔注射链脲佐菌素建立糖尿病模型。弥可保组给予弥可保混悬液0.15 mg/(kg·d)灌胃,桂枝汤组给予桂枝汤4 g/(kg·d)灌胃,正常对照组和模型对照组大鼠各给予等量生理盐水灌胃,各组均灌胃7周。采用生理参数遥测系统检测大鼠的心电图并计算心率变异性(HRV),包括时域分析的平均正常RR间期的标准差(SDNN)、相邻RR间期差的均方根(RMSSD)、每5min正常RR间期平均值的标准差(SDANN)和频域分析的低频(LF)、高频(EF)、总功率(TP)及LF/EF;检测室间隔和左心室心肌匀浆中酪氨酸羟化酶(TH)、胆碱乙酰氨基转移酶(Ch AT)、生长相关蛋白43(GAP-43)、神经营养因子(NGF)和睫状神经营养因子(CNTF)含量。结果与模型对照组比较,桂枝汤组大鼠SDNN、RMSSD和LF、HF、TP增高,而SDANN和LF/HF降低;大鼠室间隔和左心室Ch AT增加、TH/Ch AT降低,室间隔和左心室GAP-43、CNTF增加,而室间隔NGF降低(P<0.05或P<0.01)。与弥可保组比较,桂枝汤大鼠SDANN和LF/HF降低,RMSSD、LF、HF和TP升高;大鼠室间隔和左心室Ch AT、GAP-43、CNTF增加,TH/Ch AT降低(P<0.05或P<0.01)。结论桂枝汤可预防糖尿病大鼠心脏自主神经损伤,减轻自主神经重塑,其机制可能与调节交感神经和迷走神经平衡有关。nomlc nerve re Objective To observe the effect and possible mechanism of Guizhi Tang (桂枝汤) on cardiac auto- modeling of rats with diabetes. Methods Randomly divided 24 rats into normal control group, model control group, Methycobal group, and Guizhi Tang group, with 8 rats each. All groups except the normal control group adopted one-time intraperitoneal injection Streptozotocin to build diabetes model. The Methycobal group was perfused with Methycobal suspension 0. 15 rag/( kg·d). The Guizhi Tang group was given Guizhi Tang 4 g/( kg · d) for gavage. The normal control group and the model control group were given equal dose normal saline for gavage respectively. All groups were perfused for 7 weeks. Adopted physiological parameters radiotelemetry system to examine electrocardiogram and calculated heart rate variability (HRV) of rats, including standard diviation of NN intervals (SDNN), root mean square of sussessive differences (RMSSD), standard diviation average of NN intervals (SDANN) every 5 min of time-domain analysis, and low frequency ( LF), high frequency (EF) and total power (TP) and LF/EF of frequency domain analysis. Examined content of tyrosine hydroxylase (TH), Choline acetyl amino transferase (CHAT), growth-associated protein 43 (GAP-43), neurotrophic factors (NGF) and ciliary neuro- trophic factor (CNTF) in homogegate of interventricular septum and left ventricle. Results SDNN, RMSSD, LF, HF and TP of rats in the Guizhi Tang group increased while SDANN, LF/HF decreased compared with that of the model control group. ChAT increased and TH/ChAT decreased of interventricular septum and left ventricle of rats. GAP-43 and CNTF of interventricular septum and left ventricle increased while NGF of interventricular septum decreased (P 〈 0.05 or P 〈 O. 01 ).
