摘 要:目的观察补肾解毒方对辐射损伤小鼠的防护作用及其可能作用机制。方法将TLR4基因敲除(TLR4-/-)小鼠和未敲除(TLR4+/+)小鼠随机分为空白对照组、辐射模型组、中药组。适应性喂养7天后,各空白对照组、辐射模型组小鼠均给予0.2ml生理盐水,中药组给予补肾解毒方13.50g/(kg·d),连续灌胃10天,第11天除TLR4十/+和TLR4-/-空白对照组外,其余各组小鼠进行一次性60Coγ射线6Gy剂量照射,照射后第2天各组继续以上剂量灌胃14天。观察各组小鼠辐射后存活情况,记录各组小鼠辐射后第1、14、30天的体重、胸腺及脾脏质量,计算胸腺系数、脾脏系数。结果TLR4+/+小鼠辐射模型组在辐射后第2-14天共计死亡41只,中药组辐射后第7-10天共计死亡15只;TLR4-/-小鼠辐射模型组辐射后第5-13天共计死亡39只,中药组小鼠在辐射后第4~13天共计死亡38只。与同鼠种空白对照组相比,辐射模型组和中药组辐射后第1天小鼠胸腺系数、脾脏系数均有不同程度的降低(P〈0.05或P〈0.01)。TLR4+/+和TLR4-/-小鼠中药组辐射后第14、30天胸腺系数、脾脏系数较本组辐射后第l天明显升高(P〈0.05或P〈0.01)。TLR4+/+小鼠中药组辐射后第14、30天胸腺系数较辐射模型组明显升高(P〈0.05或P〈0.01);而TLR4-/-小鼠中药组与辐射模型组比较胸腺系数、脾脏系数差异均无统计学意义(P〉0.05)。结论补肾解毒方可能通过TLR4信号途径显著减轻核辐射损伤,保护免疫器官,有较理想的辐射防护作用。Objective To observe the protective effect and possible mechanism of Bushen Jiedu Fang (补肾解毒方) on mice with radiation injury. Methods Randomly divided mice of TLR4 gene knockout (TLR4 -/- ) and none knockout (TLR4 +/+ ) into blank control group, radiation model group and Chinese medicine group. Gave normal saline 0. 2 ml to mice in the blank control group and the radiation model group, and the Chinese medicine group was given Bushen Jiedu Fang 13.50 go( kg ~ d) after adaptive feeding for 7 days. Mice in all groups except the TLR4 +/+ and TLR4 -/- blank control group were given one-time 60 Co γ ray 6Gy dose exposure after 11 days of gavage. Each group continued to receive 14-day gavage of above dose the second day after exposure. Observed the survival condition of mice after radiation exposure. Recorded the body weight and the weight of thymus and spleen, on the first, 14th, and 30th day after radiation. Calculated thymus coefficient and spleen coefficient. Results TLR4 +/+ mice in the radiation model group which died 2 to 14 day after radiation exposure counted up to 41 in total, while that of the Chinese medicine group counted up to 15 mice during 7 to 10 day after radiation; TLR4 -/- mice in the radia- tion model group which died 5 to 13 day after radiation exposure counted up to 39 in total, while that of the Chinese medicine group counted up to 38 mice during 4 to 13 day after radiation; Compared to the same mice type in the blank control group, thymus coefficient and spleen coefficient of mice in the radiation group and the Chinese medicine group decreased to different degrees one day after treatment (P 〈0. 05 or P 〈0. 01 ). Thymus coefficient and spleen coeffi- cient of TLR4 +/+ and TLR4 -/- mice in the Chinese medicine group 14 and 30 day after radiation remarkably increased compared to that of this group one day after treatment ( P 〈 0. 05 or P 〈 0. 01 ).
