摘 要:目的:探讨归芪补肾颗粒对阿霉素导致的足细胞损伤的保护作用。方法:以0.5μg·mL^(-1)阿霉素复制足细胞损伤模型,再分别以低、中、高剂量归芪补肾颗粒含药血清(2%,4%,8%)干预24 h后,分别检测各组细胞nephrin、p-cadherin、desmin、α-SMA mRNA(RT-PCR法)和蛋白(Western Blot法)表达。结果:RT-PCR结果显示:与空白对照组(C组)比较,阿霉素模型组(Model)nephrin、P-cadherin mRNA表达(0.342±0.140,0.094±0.012)显著降低(P=0.000),desmin、α-SMA mRNA表达(0.517±0.096,1.242±0.198)显著增高(P=0.001,0.000);与模型组比较,归芪补肾颗粒中(M-GQBS)、高剂量组(H-GQBS)nephrin、P-cadherin mRNA表达(0.536±0.086,0.254±0.060,0.546±0.113,0.274±0.069)显著降低(P=0.007,0.005,0.001,0.000),desmin、α-SMA mRNA表达(0.393±0.084,0.819±0.067,0.375±0.084,0.764±0.125)显著增高(P=0.024,0.010,0.000,0.000)中、高剂量组之间没有差异(P>0.05)。Western Blot结果显示,model组nephrin及Pcadherin蛋白含量显著降低(P=0.000),demin、α-SMA蛋白含量显著增高(P=0.000);与Model组比较,M-GQBS及HGQBS组nephrin、P-cadherin蛋白表达显著增高(P=0.002,0.003,0.005,0.002),demin、α-SMA含量显著降低(P=0.000)。结论:归芪补肾颗粒能减轻阿霉素引起的足细胞损伤,减缓足细胞上皮-间充质转分化。OBJECTIVE To investigate protective effects of Guiqibushen Granules on podocyte damaged by Adriamycin.METHODS A model of podocyte damage induced by 0.5μg/mL Adriamycin was established,and then intervened 24 h later by low,medium,or high doses of Guiqibushen granules containing serum(2%,4%,8%).Expressions of mRNA(RT-PCR)and proteins(Western Blot)of nephrin,P-cadherin,desmin andα-SMA in each group were detected.RESULTS RT-PCR results showed that,compared with control group(C group),expressions of nephrin,P-cadherin mRNA(0.342±0.140,0.094,0.012)in Adriamycin model group(Model group)decreased significantly(P=0.000),and expressions of desmin,α-SMA mRNA(0.517±0.096,1.242±0.198)increased obviously(P=0.001,0.000).Compared with model group,expressions of nephrin and P-cadherin mRNA in medium and high doses of Guiqibushen granules(M-GQBS,H-GQBS)(0.536±0.086,0.254±0.060,0.546±0.113,0.274±0.069)decreased significantly(P=0.007,0.005,0.001,0.000),and desmin andα-SMA mRNA(0.393±0.084,0.819±0.067,0.375±0.084,0.764±0.125)increased significantly(P=0.024,0.010,0.000,0.000).No difference was observed between middle and high dose groups(P〉0.05).WB results showed that,compared with C group,expressions of nephrin and P-cadherin protein decreased significantly(P=0.000),and demin andα-SMA increased obviously(P=0.000).Compared with model group,nephrin and P-cadherin protein of M-GQBS,H-GQBS increased significantly(P=0.002,0.003,0.005,0.002),and desmin andα-SMA decreased significantly(P=0.000).CONCLUSION Guiqibushen granules can reduce podocyte damage induced by adriamycin,and relieve epithelial-mesenchymal-transition(EMT)of podocyte.
