摘 要:目的探讨宫颈癌细胞株HeLa与正常宫颈细胞株Ect1/E6E7中微小RNA(miRNA)分子表达谱的差异。方法选取HeLa细胞和Ect1/E6E7细胞各3个样本分别作为实验组和对照组,采用miRNA芯片筛选两者之间差异表达的miRNA,样品间表达变化标准以上/下调4倍作为阈值范围,并运用miRWalk软件预测差异表达miRNA可能调控的靶基因,并对这些靶基因进行信号通路的富集分析。结果在HeLa细胞中,上调超过4倍差异表达miRNA分子7个,下调超过4倍差异表达miRNA分子16个(P<0.01)。生物信息学分析发现hsa-miR-126-3p的靶基因在肿瘤通路、凋亡通路以及Wnt信号通路上出现聚集;而hsa-miR-25-5p的靶基因肿瘤通路和钙离子信号通路出现聚集;且hsa-miR-205-5p靶基因在TGF-β信号通路和EGFR信号通路出现聚集。结论宫颈癌细胞HeLa与正常宫颈细胞Ect1/E6E7之间存在差异表达miRNA分子,他们可能参与宫颈癌的发生发展机制。Objective To explore microRNA differential expression profile between cervical cancer cell line HeLa and normal cervical epithelial cell line Ectl/E6ET. Methods Three samples of HeLa cell line and another three samples of Ectl/E6E7 cell line were selected as experimental group and control group, respectively. MicroRNA profiles in these samples were screened by microarray. The fold change of threshold values used to screen up-and down-regulated microRNAs was more than 4. The target genes of differential expressed miRNA were predicted by miRWalk software and the related signal pathways were analyzed by enrichment analysis. Results Seven molecules expressing differential microRNA were up-regulated and sixteen molecules expressing differential microRNA were down-regulated in HeLa cell line, compared with Ectl/E6E7 cell line, there were statistically significant differences (P〈0.01) . Bioinformatic analysis showed that the target genes of hsa-miR-126-3p were enriched in cancer pathway, apoptosis pathway, and Wnt signal pathway; the target genes of hsa-miR-25 -5p were enriched in cancer pathway and calcium signaling pathway; the target genes of hsa-miR-205-Sp were enriched in TGF-β signal pathway and EGFR signal pathway. Conclusion These differentially expressed microRNA in cervical cancer cell line HeLa and normal cervical epithelial cell line Ectl/E6E7 might play important roles in occurrence and development of cervical cancer.
