摘 要:目的研究红参提取物(HSI)对人脐静脉内皮细胞分泌一氧化氮(NO)、内皮素-1(ET-1)与6-酮前列腺素F_(1α)(6-K-PGF_(1α))的影响。方法体外培养人脐静脉内皮细胞株EA.hy926,随机分为5组:空白组、对照组、3个剂量(40,20,10 mL·L-1)实验组,将细胞传代培养后,分别用格里斯(Griess)试剂盒检测细胞内外的NO含量;用酶联免疫吸附测定法检测细胞上清液中的ET-1、内皮型一氧化氮合酶(eNOS)、环氧化酶-1(COX-1)以及6-酮-前列腺素F_(1α)(6-K-PGF_(1α))的含量。结果小中2个剂量HSI均可显著增加细胞内外的NO含量(P<0.05),3个剂量均可提高eNOS表达水平(P<0.05);大中2个剂量组可以减少细胞上清液中ET-1含量(P<0.05),而对COX-1以及6-K-PGF_(1α)的含量无明显影响(P>0.05)。结论 HSI可通过促进NO合成与释放,降低ET-1含量,发挥舒张血管作用。Objective To investigate the mechanism of Red Ginseng extraction( Hongshen injection,HSI) on nitric oxide( NO),endothelin-1( ET-1) and 6-keto-prostaglandin( 6-K-PGF_(1α)) released by human umbilical vein endothelial cells. Methods Cultured human umbilical vein endothelial cells EA. hy926 which have been randomly divided into five groups,blank group,control group,and three doses( 40,20,10 m L · L-1) test groups. Tested of the content of NO both by using Griess kit. Then the content of ET-1,endothelial nitric oxide synthase( eNOS),cyclooxygenase-1( COX-1) and 6-K-PGF_(1α)in the cell supernatant were detected by enzyme linked immunosorbent assay( ELISA). Results Low and middle dose of HSI could increase the content of NO( P〈0. 05). Three dose HSI enhance the eNOS expression level( P〈0. 05). Middle and high dose HSI reduce the content of ET-1 in cell supernatant which is similar with the effect of Sildenafil. The20,40 m L·L-1HSI can reduce the content of ET-1 in cell supernatant( P〈0. 05),but has no obvious effect on the content of COX-1and 6-K-PGF_(1α)( P〉0. 05). Conclusion HSI can relax the blood vessel by promoting the NO synthesis and release,reducing ET-1,the effect is based on the NO pathway by increasing the expression of e NOS but not on the prostacyclin I2( PGI2) pathway.
