摘  要:[目的]通过研究γ-L-谷氨酰-S-烯丙基-L-半胱氨酸(GSAC)对大鼠肝星状细胞(HSC-T6)线粒体功能的影响,探讨GSAC对肝纤维化的抑制作用。[方法]利用微板法测定GSAC对培养24 h细胞线粒体代谢甲基噻唑基四唑(MTT)的影响,以罗丹明123作为染料,通过流式细胞仪检测线粒体膜电位。[结果]GSAC具有抑制HSC-T6线粒体代谢MTT的作用,IC50值为0.835 mg/ml;GSAC引起线粒体膜电位崩溃,所有GSAC处理组(0.2、0.4与0.8 mg/ml)罗丹明123荧光强度极显著高于对照组。[结论]GSAC可破坏HSC-T6细胞线粒体功能,提示其对肝纤维化具有抑制作用。[ Objective] The research aimed to explore inhibition effect of γ-L-glntamoyl-S-allyl-L-cysteine (GSAC) on liver fibrosis by studying the influence of GSAC on the mitoehondrial function of rat hepatic stellate cells (HSC-T6). [ Method J After 24 h culture in different concentrations of GSAC, methyl thiazolyl tetrazolium (MTT) metabolism was tested, with the median inhibitory concentration ( IC50) value obtained in a microplate assay, and mitochondrial membrane potential (MMP) was tested in a flow cytometer assay with Rhodamine 123 as indicator. [Result]GSAC inhibited MTr metabolism, with IC50 =0.835 mg/ml, and GSAC inhibited MMP, the fluorescence intensity in GSAC treated groups (0.2, 0.4 and 0.8 mg/ml) significantly increased compared to the control. [ Conclusion] GSAC damages mitochondrial function of HSC-T6 cells, which suggests that GSAC has an inhibitory effect on liver fibrosis.
