广西甘蔗杆状病毒的分子检测及分布调查

摘　　要：【目的】明确广西蔗区甘蔗杆状病毒病的发生及分布情况,了解不同甘蔗品种(品系)中甘蔗杆状病毒(Sugarcane bacilliform virus,SCBV)的侵染情况,为针对性地开展甘蔗杆状病毒病的防控提供理论依据。【方法】20122014年从广西14个地市的主要甘蔗种植区采集表现褪绿斑点、斑驳及无症状甘蔗叶片样品169份,同时从南宁的甘蔗育种和示范基地采集12个甘蔗品种(品系)叶片样品,采用杆状DNA病毒属(Badnavirus)通用检测引物Badna FP/RP对采集的样品进行PCR检测和测序分析。【结果】采自广西各地的169份样品中有105份为SCBV阳性,检出率达62.1%;样品中糖蔗与果蔗的SCBV阳性率相差不明显,分别为63.5%和58.1%;表现斑点或花叶的甘蔗叶片中SCBV的检出率较高,部分没有明显症状的甘蔗样品也检测到SCBV的存在。12个品种(品系)中包括新台糖系列、桂糖32、桂糖40、粤甘24、粤糖60和福农38等在内的7个甘蔗品种(品系)均检测到SCBV的核酸序列。【结论】目前广西蔗区存在SCBV侵染,且已经广泛分布。[Objective]The occurrence and distribution of sugarcane bacilliform virus（SCBV） in Guangxi sugarcane area were investigated to understand infection situation of different sugarcane varieties,in order to provide theoretical basis for controlling SCBV diseases.[Method]A total of 169 sugarcane leaf samples with chlorosis spots and mottle,and without symptoms were collected from main sugarcane areas in 14 cities of Guangxi during 2012-2014.Meanwhile,leaf samples of 12 sugarcane varieties were collected from Nanning sugarcane breeding and demonstration base.Badnavirus-specific universal primers（Badna FP/RP） were used to detect collected samples by PCR amplification and sequence analysis.[Result]The results showed that,among 169 tested samples,SCBV was found in 105 samples,thus positive rate were up to 62.1%,there was no significant difference in SCBV positive rates between sugar cane（63.5%） and fruit cane（58.1%）.Samples with spots or mosaic symptoms got higher detection rate,and partial samples with no obvious symptoms also were found to be SCBV positive.Among 12 sugarcane varieties collected from sugarcane breeding and demonstration base of Nanning,there were 7 sugarcane varieties viz.,ROC series,Guitamg 32,Guitamg 40,Yuegan 24,Yuetang 60,Funong38 were found to contain SCBV sequence.[Conclusion]At present,SCBV has been widely dispersed in sugarcane growing area of Guangxi province.
