摘  要:目的:阐明桂枝汤对大鼠变应性鼻炎鼻黏膜上皮细胞AQP5表达及c AMP/PKA-CREB信号通路的影响。方法:通过卵清蛋白全身致敏,以及局部激发的方法,建立变应性鼻炎大鼠的动物模型,灌胃给予桂枝汤低、高剂量(4.2、8.4g/kg)14d,通过免疫组织化学染色、Western Blot和荧光定量PCR检测AQP5、p-CREB(Ser133)的表达。结果:桂枝汤高、低剂量均能明显减轻鼻黏膜组织病理改变,增加AQP5、p-CREB(Ser133)蛋白和AQP5m RNA的表达。结论:桂枝汤对变应性鼻炎具有一定的治疗作用,该作用机制可能与调节AQP5表达及c AMP/PKACREB信号通路有关。Objective:To explore the effects of Guizhi decoction on the expression of AQP5 and cAMP/PKA-CREB signaling pathway in the epithelial cells of nasal mucosa of allergic rhinitis rats.Methods:A rat model of allergic rhinitis was established by ovalbumin systemic and local sensitization.Rats were given the low dose of Guizhi Decoction(4.2g/kg) and high dose of Guizhi Decoction(8.4g/kg) by gavage for 14 days.The expressions of AQP5 and p-CREB(Ser133) were detected by immunohistochemistry,Western Blot and fluorescence quantitative PCR.Results:Both of the low dose of Guizhi Decoction and the high dose of Guizhi Decoction could significantly induce the pathologic changes of nasal mucosa tissue.Meanwhile,they could increase the protein and gene expressions of AQP5 and p-CREB.Conclusion:Guizhi Decoction has therapeutic effect on allergic rhinitis rats,and the mechanism may be related to regulate the expression of AQP5 and cAMP/PKA-CREB signal pathway.
