摘　　要：目的:观察温阳补肾方对兔激素性股骨头坏死组织RANKL/RANK/OPG通路的影响。方法:选取普通级健康新西兰大白兔136只,随机分为正常组,模型组,中药低、中、高剂量组。干预8周后运用Western-blot检测OPG、RANK、RANKL蛋白表达。结果:中药各剂量组OPG的表达量均高于模型组(P<0.05,P<0.01),而RANK、RANKL的表达量低于模型组(P<0.01)。随用药剂量的增加,OPG蛋白相对表达量逐渐升高,RANK、RANKL蛋白相对表达量逐渐降低。结论:温阳补肾方能激活RANKL/RANK/OPG通路,提高激素性股骨头坏死兔股骨头组织中OPG的表达,抑制RANK、RANKL的表达,从而刺激成骨细胞与破骨细胞的功能及活性,这可能是其有效防治股骨头坏死的机制之一。Objective：To investigate effects of Wenyang Bushen Formula on the pathway of RANKL/RANK/OPG in the rabbits with femoral head necrosis induced by steroid.Methods：136 New Zealand rabbits were randomly divided into 5 groups：normal group,model group,low dose of Wenyang Bushen Formula group,moderate dose of Wenyang Bushen formula group and high dose of Wenyang Bushen Formula group.The expressions of OPG,RANK,and RANKL proteins in the femoral head tissues were determined by western blotting,respectively.Results：The expression of OPG protein in all TCM treatment groups was higher than that in model group（P〈0.05,P〈0.01）,the expressions of RANKL and RANK proteins in all TCM treatment groups,however,were both lower than those in model group（P〈0.01）.Conclusion：Wenyang Bushen Formula could up-regulate the expression of OPG and inhibit the expressions of RANKL and RANK in the rabbits femoral head necrosis tissue induced by steroid.We concluded that Wenyang Bushen Formula could activate the RANKL/RANK/OPG signaling pathway to stimulate the activation of osteoblast and osteoclast,which might be one of mechanisms for the prevention and treatment of femoral head necrosis.
