棘霉素对伊马替尼敏感及耐药K562细胞ERK、AKT和SPK表达的影响

摘  要:目的比较棘霉素对伊马替尼敏感及耐药K562细胞ERK、AKT和SPK表达的影响。方法 MTT法检测细胞存活率,Western blot检测ERK和AKT的表达,同位素掺入检测SPK的活性。结果棘霉素对K562和耐伊马替尼细胞的IC50分别为0.82mg/L和0.87mg/L;棘霉素对ERK和AKT均有不同程度的抑制作用;耐药K562细胞中SPK的表达远高于敏感细胞,且棘霉素对耐药细胞中的SPK活性有显著抑制作用。结论棘霉素对敏感及耐药K562细胞均有较强的抑制作用,其机制与抑制ERK、AKT和SPK的作用有关。Objective This study was aimed to explore the effect of echinomycin on the K562 and K562/Ima cells.Methods In experiments,the cells proliferation was detected by MTT assay.ERK,AKT and SPK were examed by Western blot assay,TLC in K562 and K562/Ima cells,respectively.Results The results revealed the IC50 of echinomycin in K562 and K562/Ima were 0.82 mg/L and 0.87 mg/L,respectively.The ERK and AKT were inhited by echinomycin in K562 and K562/Ima.The expression of SPK was up-regulated in K562/Ima cells,and echinomycin on SPK activity in resistant cells had obvious inhibitory effect.Conclusion Echinomycin can inhibits the proliferation of K562 and K562/Ima cells,and its mechanism may be related to inhibition of ERK,AKT and SPK.
