摘  要:2003年在荷兰禽流感爆发期间,感染的89个人中,88例患有结膜炎,1例因肺炎死亡。本研究综述比较在致死性病例(FC)病毒与结膜炎病例(CC)病毒间的生物学差异,以探究病毒的病原性与发病机制。显著的差异在于FC病毒主要结合于下呼吸道的终末端,包括肺泡。攻击1型、2型肺细胞与肺泡巨噬细胞。用FC病毒接种小鼠,其肺病毒滴度比CC病毒高1 000倍,且播散到脾、肝、肾及脑。FC病毒的血凝素(HA)基因是病毒全身性播散的决定基,FC病毒在碱性多聚酶2(PB2)上627位的赖氨酸(E627K)是毒力的主要决定基。Among the 89 cases in the outbreak of avian influenza H7N7 in the Netherlands in 2003,88 infected persons suffered from mild illnesses(conjunctivitis)and 1died of pneumonia.This review compared the biological differences of the viruses between the fatal cases(FCs)and conjunctivitis cases(CCs)in order to explore the pathogenicity of FC viruses.A marked difference was that the FC viruses attached and bound to lower respiratory tract including pulmonary alveoli,pneumocytes and alveolar macrophages.In the lungs of mice inoculated with the FC virus,the virus titers were 1000 folds higher than CC virus,and were detected in spleen,liver,kidneys and brain.Hemagglutinin(HA)gene of the FC viruses was a determinant of systematic distribution of the viruses.The lysine at position 627(E627K)of basic polymerase 2(PB2)of the FC viruses was the major determinant of the viruses′virulence.
