摘 要:对引起四川省某水库网箱养殖的加州鲈发生以体表出血、皮肤溃疡、肝肾出血和肿大为典型症状的疾病进行病原菌的分离、鉴定和致病性研究。从患病加州鲈病变组织分离纯化得到1株病原菌(LB140811),对其进行了形态特征、理化特性等表型生物学检验和回归试验,并通过16SrDNA和gyrB基因测序与系统发育分析,以及毒力基因act和aerA的PCR扩增进行分离株的分子生物学鉴定,同时开展了病原菌的致病性和药物敏感性研究。根据该病原菌的表型、分子生物学特性和进化关系,判定其为气单胞菌属的维氏气单胞菌,且回归试验证实该菌株LB140811为此次加州鲈发病的病原菌。组织病理学观察发现,该病原菌对加州鲈的多个组织器官都造成了明显的病理损伤,尤其是肝、肾、肌肉的损伤较为严重,表现为明显的变性、坏死以及炎性细胞浸润。药敏试验结果表明,18种抗菌药物中,该病原菌仅对阿奇霉素敏感,对氨苄西林、氧氟沙星等14种药物耐药。In order to study the isolation, identification and pathogenicity o{ the pathogen LB140811 isolated from the diseased Micropterus salmoides which was {armed in reservoir cages of Sichuan province and characterized by the surface bleeding, cutaneous ulcers, visceral organs swelling,congestion or bleeding. The morphologic structure, physiological and biochemical tests, artificial infection experiment was conducted,followed by identification with 16S rDNA and gyrB genes,and detection of virulence genes act (cytotoxic enterotoxin) and aerA (aerolysin) by PCR amplication. The pathogenicity and drug sensitivity test of the isolate was also carried out. The re- suits showed that the pathogen LB140811 was identified as Aeromonas veronii according to the phenotypic biology,molecular biological characteristic and phylogenetic trees analysis,which was confirmed as the pathogen caused the diseased Micropterus salmoides by artificial infection exper- iment. The histopathological observation indicated that the pathogen caused Micropterus salmo- ides obvious pathological damage on some tissues and organs, especially in liver, kidney and mus- cle,and the main pathologic lesions were tissues degeneration and necrosis as well as infiltration of the inflammation cells. Moreover, the results of drug sensitivity tests showed that among the 18kinds of antibiotics, the pathogen was only sensitive to azithromycin, while resistant to ampicillin, ofloxacin and other 14 kinds of drugs.
