摘 要:从猪肺泡巨噬细胞中克隆了猪干扰素调节因子1(interferon regulatory factors 1,IRF1)基因,并构建了真核表达载体pCAGGS-HA-IRF1。将该质粒转染PK-15后通过Western-blot确定IRF1蛋白在细胞内能够高效表达。通过体外抗病毒试验发现,过表达IRF-1能够显著抑制H1N1流感病毒和猪水泡性口炎病毒的复制。进一步通过双荧光报告系统发现,IRF1显著促进polyI:C诱导的猪IFN-β、ISRE和NF-κB启动子的活性,说明IRF1可能通过增强细胞的天然免疫应答进而发挥抗病毒功能。In order to study the anti-viral effects and immune-modulatory actions of IRF1 ,porcine IRF1 gene was cloned firstly from porcine pulmonary alveolar epithelial cells and inserted into the plasmid of pCAGGS (designed as pCAGGS-HA-IRF1), pCAGGS-HA-IRF1 was transfected into PK-15 cells and subseaquently Western-blot corfirmed the highly effective expression of IRF1 protein. In vitro antiviral tests showed that over-expression of IRF-1 can significantly inhibit the replication of H1N1 influenza virus and swine vesicular stomatitis virus. Further studies by dual luciferase reporter system showed that IRF1 significantly promoted promoter activities of polyI :C-induced porcine IFN-~, ISRE and NF-~:B, indicating that IRF1 may thus play the anti-viral role by enhancing cellular innate immune response. This study will be contributed to further explore the antiviral effects and immune-modulatory functions of porcine IRF1.
