摘 要:本试验旨在研究不同剂量铝对小鼠F9畸胎瘤细胞周期、凋亡和DNA-蛋白质交联(DNA-protein crosslinks,DPC)的影响。不同水平的氯化铝(0、50、100、200μmol/L,Ⅰ~Ⅳ组)作用F9细胞24h后,采用MTT法观察细胞的增殖毒性,流式细胞术和荧光染色法分别研究细胞周期和凋亡的变化,荧光比色法研究DPC含量的变化。结果显示:与Ⅰ组相比,各剂量组细胞的增殖活力、谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)活性呈不同水平的降低,S期细胞数量的百分比、细胞凋亡率、乳酸脱氢酶(LDH)释放水平和丙二醛(MDA)含量不同程度的升高,存在剂量-效应相关。结果表明:过量的铝能够打破F9细胞氧化抗氧化的平衡,对细胞DNA产生损伤。The aim of this in vitro study was to evaluate the effects of aluminum on cell cycle, ap- optosis and DNA-protein crosslink (DPC) in F9 embryonic carcinoma cells exposed to 0,50,100, and 200/lmol/L A1C13 ( Ⅰ ,Ⅱ , Ⅲ and Ⅳ group). After incubation for 24 h,the proportions of via- ble cells were measured by MTT. The percentages of cell cycle and apoptosis were measured by flow cytometry and fluorescence staining,respectively. Fluorometric spectrophotometry was used to determine the levels of DPCs in the F9 cells. Cell vability decreased in the groups treated with 100μmol/L and above when compared with the I group. Compared with I group,the cell viabil- ity,GSH-Px and SOD activities were decreased,while the contents of LDH, MDA, percentage of S phase,and ratio of apoptosis were increased in dose-dependent. Excessive aluminum could disrupt the state of oxidant/antioxidant,which induces DNA damage in F9 cells.
