桔梗乙酸乙酯提取段对酒石酸泰乐菌素在小鼠血浆和肺组织中药动学的影响

摘　　要：采用高效液相色谱法(HPLC)测定不同时间点小鼠血浆和肺组织中酒石酸泰乐菌素的浓度,运用PKsolver程序及SPSS13.0统计软件处理和分析数据,计算相关药代动力学参数。结果显示:与对照组相比,乙酸乙酯组半衰期(t1/2)变短,Cmax增大、,药时曲线下面积(AUC)增加、Tmax与平均滞留时间(MRT0-inf_obs)缩短,而表观分布容积(Vz/F_obs)与消除率(Cl/F_obs)变小;肺组织分布图显示药物浓度高于对照组。结果表明:桔梗乙酸乙酯提取活性段对酒石酸泰乐菌素在小鼠体内的药动学特征有较大影响,表现为加快了酒石酸泰乐菌素的吸收速度,并增加了吸收程度;但药物在体内分布变窄,却增加了其在肺中的药物浓度。We used tylosin tartrate as the target drug to investigate the influence of ethyl acetate extract of Platycodon grandiflorum roots on the pharmacokinetics and lung tissue distribution character of tylosin tartrate in mice, after oral administration. The method is a high performance liquid chromatography was used for determining the mice plasma and tissue concentrations of tylosin tartrate at different time points, then the pharmacokinetic data was calculated by Pkslover program and SPSS13.0 software and analyzed by non compartment model. Compared with con- trast group, the main pharmacokinetics parameters in test group were changed as follows：half-life time（t1/2 ）became shorter, the mean residence time（MRT）and peak time were decreased, and the same wih numerical value of apparent volume of distribution （Vd）and clearance （C1）, however peak concentra- tion and area under the curve（ AUC） were increased; drug concentration was higher than that in the con- trast group in lungs. The result showed that ethyl acetate extract of Platycodon grandiflorum roots could influence the pharmacokinetic parameters of tylosin tartrate in healthy mice significantly,and the influence included promoting the speed and degree of the absorption, narrowing the scope of the distribu- tion of drug in the body,and advancing the drug concentration in the lung.
