摘  要:目的观察不同浓度葡萄籽提取物原花青素(GSPE)对长波紫外线(UVA)诱导人皮肤成纤维细胞表达转化生长因子-β(TGF-β)Ⅱ型受体和Smad7蛋白的影响。方法本实验选用体外培养的人皮肤成纤维细胞株(HSFs)为研究对象,实验主要分3组,空白对照组,单纯光照组(UVA照射人皮肤成纤维细胞),照光加药组(UVA照射细胞后予不同浓度GSPE干预)。应用免疫细胞化学技术检测UVA照射前、后及不同浓度GSPE干预后对皮肤成纤维细胞TGF-βRⅡ和Smad7蛋白表达的情况。结果与空白对照组相比,单纯UVA光照组人皮肤成纤维细胞TGF-βRⅡ蛋白表达下降(P<0.05),Smad7蛋白表达升高(P<0.05);与单纯光照组相比,UVA照射后经25μg/m L、50μg/m L和100μg/m L浓度GSPE处理后,成纤维细胞TGF-βRⅡ蛋白表达水平均升高(P<0.05),Smad7蛋白表达水平均下降(P<0.05),且于50μg/m L和100μg/m L GSPE处理后,成纤维细胞TGF-βRⅡ和Smad7蛋白表达量均接近正常水平(P>0.05)。结论适当浓度GSPE可通过显著上调TGF-βRⅡ蛋白水平以及下调Smad7蛋白水平的表达来对抗UVA抑制TGF-β/Smad信号通路的传导,进而减轻UVA所致细胞外基质的降解,延缓了皮肤光老化的进程。Objective To explore the photoprotection and the molecular mechanisms of grape seed proanthocyanidin extract(GSPE) on UVA irradiation-induced extracellular matrix by studying the TGF-β/Smad signal transduction. Methods Cultured human skin fibroblasts were treated with three groups. HSFs induced by UVA irradiation were pure light group, and then cultured with different concentrations of GSPE were GSPE intervention group. The third group is control group. The protein levels of TGF-β RⅡ and Smad7 in cultured fibroblasts were detected by immunocytochemistry method. Results Comparing with the control group, the protein levels of TGF-β RⅡ in pure light group were markedly decreased, and conversely the protein expression of Smad7 were obviously increased(P〈0.05). The protein levels of TGF-β RⅡ were increased after treated with25 μg/m L, 50 μg/m L and 100 μg/m L GSPE(P〈0.05), and the protein expression of Smad7 were simultaneously decreased(P〈0.05).After treated with 50 μg/m L or 100 μg/m L GSPE, the protein expression of TGF-β RⅡand Smad7 were close to normal in GSPE intervention group(P〉0.05). Conclusion GSPE in appropriate concentration could resist the UVA-damaged in the fibroblasts through regulating the expression of related molecules in the TGF-β/Smad signaling pathway, relieve the degradation of extracellular matrix and delay the process of the skin photoaging.
