摘 要:目的了解野生鸟类新城疫病毒(Newcastle disease virus,NDV)感染情况,分析NDV的基因型与遗传变异特点。方法采用HA试验和RT-PCR检测野生鸟类新城疫病毒感染情况,对新城疫病毒分离株的融合蛋白基因(F基因)和血凝素-神经氨酸酶基因(HN基因)进行遗传进化分析,了解F和HN基因相关分子特性。结果 7株NDV的F基因序列分析表明显示其编码区核苷酸为1 662bp,编码553个氨基酸。F基因碱性裂解位点序列为112G-K/RQ-G-R-L117,均符合新城疫弱毒特征,与Ⅱ类NDV代表株核苷酸同源性为93.5%~99.9%。M46与M52为Ⅱ类Ⅱ亚型,其余5株为Ⅱ类Ⅰ亚型。HN基因序列分析显示,M46、M52编码区核苷酸为1 734bp,编码577个氨基酸,其余5株编码区氨基酸为1 851bp,编码616个氨基酸,与Ⅱ类代表毒株核苷酸同源性为92%~99.6%。结论分离的7株新城疫病毒在基因上均符合弱毒特点。其中有5株为Ⅱ类Ⅰ型,2株为Ⅱ类Ⅱ型。Objectives To determine the prevalence of the Newcastle disease virus(NDV)in birds and to analyze the characteristics of NDV,including its subtypes and genetic mutations. Methods The prevalence of NDV in birds was determined with a hemagglutination(HA)test and RT-PCR,and genetic evolution of the F and HN genes of NDV strains was analyzed in order to analyze the molecular characteristics of the F and HN genes. Results Sequencing of the F gene in seven NDV strains indicated that the gene had 1,662 nucleotides,corresponding to 553 amino acids,in its coding region.The motif of the cleavage site was 112G-K/R-Q-G-R-L117,which is consistent with the characteristics of avirulent NDV strains.Analysis of the homology of the F gene revealed that its sequence was 93.5%to 99.9% similar to the sequence of that gene in Class II reference strains.The strains M46 and M52were Class II genotype 2viruses but the other five strains were Class II genotype 1viruses.The HN gene of strains M46 and M52contains a 1 734 bp open reading frame and it codes for 577 amino acids.In contrast,the HN gene of the other 5strains contains a 1 851 bp open reading frame.Analysis of homology revealed that the HN gene in the seven isolates was 92%-99.6% similar to the same gene in Class II reference strains. Conclusion Phylogenetic analysis of the F gene and HN gene revealed that seven isolates could be categorized as lentogenic viruses;five strains were grouped into genotype 1in Class II and the other two were grouped into genotype 2in Class II.
