摘  要:目的观察不同剂量亚砷酸钠(NaAsO2)对PCI2细胞(具有神经元特性的大鼠嗜铬瘤细胞)色素上皮衍生因子(PEDF)和凋亡相关因子mRNA表达的影响。方法选择PC12细胞作为研究对象,给予不同剂量的NaAsO2[0(对照组)、2、5、10μmol/L]染毒24h,使用Real-time荧光定量PCR检测不同剂量NaAsO2染毒后PC12细胞的PEDF及其凋亡相关因子Bax和Bcl-2mRNA的表达水平。结果PEDFmRNA表达组间比较差异有统计学意义(F=102.28,P〈0.05),其中2、5、10μmol/LNaAsO2染毒组的PEDFmRNA表达(0.70±0.07、0.33±0.04、0.23±0.10)均低于对照组(1.15±0.11,P〈0.05)。BaxmRNA表达组间比较差异无统计学意义(0、2、5、10μmoVL组分另0为0.95±0.12、0.80±0.11、0.88±0.11、1.01±0.11,F=2.01,P〉0.05);Bcl-2mRNA表达组间比较差异有统计学意义(F=19.87,P〈0.05),其中2、5、10μmol/LNaAs02染毒组Bcl-2mRNA表达(0.65±0.03、0.49±0.04、0.57±0.09)均低于对照组(0.95±0.11,P均〈0.05);Bax/Bcl-2mRNA表达比值组间比较差异有统计学意义(F=8.35,P〈0.05),其中5、10μmol/LNaAsO2染毒组的Bax/Bcl-2mRNA表达比值(1.80±0.72、1.82±0.36)高于对照组(1.02±0.24,P均〈0.05)。结论NaAsO2可能通过降低PC12细胞PEDFmRNA的表达来增加凋亡相关因子Bax/Bcl-2mRNA表达.继而引起细胞凋亡。Objective To observe the effects of different levels of sodium arsenite (NaAsO2) on mRNA expression of pigment epithelium-derived factor (PEDF) and apoptosis-related factors in PC12 cells (rat neuron properties pheochromocytoma). Methods PC12 cells were treated with different levels of NaAsO2 [0 (control group), 2, 5, 10 μmol/L] for 24 hours. The mRNA expression of PEDF and apoptosis-related factors (Bax, Bcl-2) were detected by real-time quantitative PCR. Results There were significant differences in the mRNA expressions of PEDF between the 4 groups (F = 102.28, P 〈 0.05), the mRNA expressions of PEDF in the group of 2, 5, 10 μmol/L (0.70 ± 0.07, 0.33 ± 0.04, 0.23 ± 0.10) was lower than that of control group (1.15 ±0.11, P 〈 0.05); there were no significant differences in the mRNA expressions of Bax between the 4 groups (0, 2, 5, 10 μmol/L groups: 0.95 ± 0.12, 0.80 ± 0.11, 0.88 ± 0.11, 1.01 ± 0.11, F = 2.01, P 〉 0.05); there were significant differences in the mRNA expressions of Bcl-2 between the 4 groups (F = 19.87, P 〈 0.05), the mRNA expressions of Bcl-2 in the group of 2, 5, 10 μmol/L (0.65 ± 0.03, 0.49 ± 0.04, 0.57 ± 0.09) were lower than that of control group (0.95 ± 0.11, all P 〈0.05); there were significant differences in the mRNA expressions of Bax/Bel-2 between the 4 groups (F = 8.352, P 〈 0.05), the mRNA expressions of in the group of 5, 10 μmol/L (1.80 ± 0.72, 1.82 ± 0.36) were higher than that of control group (1.02± 0.24, all P 〈 0.05). Conclusion NaAsO2 may increase the expression of apoptosis-related factorsBax/ Bcl-2 mRNA by decreasing the expression of PEDF mRNA in PC12 cells, leading to apoptosis in PC12 cells.
