摘 要:为研究运输应激对动物行为、内分泌以及脑内神经型一氧化氮合酶(Neuronal nitric oxide synthase,nNOS)表达的影响,采用摇床加制动束缚模拟公路运输途中的摇晃、拥挤和限制活动等因素,建立大鼠运输应激模型(35℃,120r/min,应激2h)。将18只大鼠随机分为对照组、应激组和应激恢复组,采用旷场试验评定大鼠行为反应,放免法测定大鼠血清皮质酮含量,免疫组化法检测大鼠下丘脑视上核(Supraoptic nucleus,SON)中nNOS的表达。结果显示,与对照组相比,应激组大鼠的自主活动明显减少(P<0.01),血清皮质酮(Corticosterone,CORT)含量显著增加(P<0.01),SON中nNOS的平均光密度值显著上升(P<0.01),应激恢复组大鼠的自主活动次数和血清皮质酮含量恢复至正常水平,而SON中nNOS的平均光密度值进一步上升。上述结果表明,运输应激对大鼠情绪、行为和神经内分泌有显著影响,24h后行为及内分泌状况改善,而神经递质的改变则需更长时间恢复。In order to investigate the impact of transport stress on behavior,endocrine and nNOS in the brain of animals, the rats were subjected to 35 ℃, 120 rpm for 2 hours on a shaker to simulate the condition of road transport. Eighteen rats were randomly divided into three groups: the control group, the transport stress group (TS group), and the transport stress recovery group (TS recovery group). Behavior of rats was assessed by open-field test. Serum corticosterone (CORT) was detected using radioimmunoassay. Expression of nNOS in the supraoptic nucleus (SON) was examined by immunohistochemical technique. After transport stress, the autonomic activities of rats were decreased (P〈0.01) and serum CORT was increased (P〈0.01), significantly. Moreover, a significant increase in nNOS expression was detected in the SON of rats in the TS group (P〈0.05) compared to the control group. Notably,the rat autonomic activities and serum CORT returned to normal level in 24 hours. However, the expression of nNOS in the SON increased further in the TS recovery group. These results suggested that transport stress has a remarkable influence on mood, behavior, endocrine and neuronal system of rats. Although behavior and endocrine condition recovered after 24 hours, the neuronal transmitter called for a longer period to recover.
