大七气汤单用及联合顺铂对SKOV-3细胞移植瘤Survivin与Caspase-3表达的影响

摘　　要：目的探讨大七气汤加减方(姜黄、三棱、莪术、半枝莲等)单用及联合顺铂对SKOV-3细胞移植瘤Survivin与Caspase-3表达的影响。方法将SKOV-3细胞移植瘤造模成功的40只荷瘤鼠随机分为模型对照组、大七气汤低、高剂量组、顺铂组、顺铂联合大七气汤组,每组8只,给药21 d后处死所有荷瘤鼠,称瘤重,测瘤体积,以逆转录-聚合酶链反应法和蛋白质印迹法检测瘤组织Survivin、Caspase-3 m RNA及蛋白的表达。结果各用药组瘤重和瘤体积均小于模型对照组(P<0.01);逆转录-聚合酶链反应和蛋白质印迹法检测结果显示中药组Survivin m RNA及蛋白表达明显低于模型对照组(P<0.01),且大七气汤低剂量组与DDP顺铂组比较差异有统计学意义(P<0.05),且DDP联合大七气汤组表达最低;用药组Caspase-3 m RNA和蛋白表达明显高于模型对照组(P<0.01),且大七气汤低剂量组与顺铂组比较差异有统计学意义(P<0.05),且顺铂联合大七气汤组表达最高。结论大七气汤加减方能够下调Survivin和上调Caspase-3的表达来达到抑制肿瘤细胞生长和促进肿瘤细胞凋亡,且大七气汤联合顺铂对SKOV-3细胞移植瘤治疗具有增效作用。AIM To explore the effects of single modified Daqiqi Decoction（ Rhizoma curcumae longae,Sparganii Rhizoma,Curcumae Rhizoma,Scutellariae barbatae Herba,etc.） and combined with DDP on Survivin and Caspase-3 expressions of human ovarian cancer cell line SKOV-3. METHODS A model of SKOV-3 cell transplanted tumor was established and forty mice with tumor were divided into model control group,Daqiqi Decoction low-,high-dose groups,DDP group and DDP combined with Daqiqi Decoction group,eight mice in each group.After successive administration for 21 d,the weights of tumor and volume were measured,and the expressions of Survivin,Caspase-3 m RNA and protein were detected by reverse transcription polymerase chain reaction（ PCR）and Western blot method. RESULTS Each group＇s tumor weight and volume were less than those of the model control group（ P〈0. 01）. The detection results of reverse transcription PCR and Western blot showed that the expressions of Survivin m RNA and protein in treatment groups were significantly lower than those of the model group（ P〈0. 01）. There was no statistical difference as compared with the DDP group（ P〈0. 05）,the DDP combined with the Daqiqi Decoction group was the lowest,and the expressions of Caspase-3 m RNA and protein in the test group were significantly higher than those in the model group（ P〈0. 01）. There was no statistical difference as compared with the DDP group（ P〈0. 05）,and the DDP combined with Daqiqi Decoction group was the highest.CONCLUSION The modified Daqiqi Decoction can achieve the inhibition of tumor cell growth and promotion of apoptosis of tumor cells by reducing the expression of Survivin and raising the expression of Caspase-3. It also has synergistic effect in the combination with DDP on SKOV-3 cell transplantation tumor.
