摘  要:目的建立HPLC波长切换法同时测定复方苁蓉散(肉苁蓉、淫羊藿、黄精)中松果菊苷、毛蕊花糖苷和淫羊藿苷的含有量。方法复方苁蓉散50%甲醇提取液分析采用Agilent HC-C18色谱柱(250 mm×4.6 mm,5μm),以乙腈(A)-0.1%磷酸溶液(B)为流动相,梯度洗脱;体积流量为1.0 m L/min;检测波长为330 nm(0~20 mim,松果菊苷和毛蕊花糖苷)和270 nm(20~30 mim,淫羊藿苷),柱温30℃。结果松果菊苷、毛蕊花糖苷和淫羊藿苷分别在142.4~2 848 ng(r=0.999 5)、19.92~398.4 ng(r=0.999 8)和19.60~392.0 ng(r=0.999 9)范围内,与峰面积呈良好的线性关系,平均回收率(n=6)分别为99.03%、99.97%、99.50%。结论该方法简便、准确、可靠,可用于复方苁蓉散中上述3种成分的质量控制。AIM To establish an HPLC wavelength switching method for simultaneously determining the contents of echinacoside,verbascoside and icariin in Compound Congrong Powder( Cistanche deserticola Y. C. Ma,Epimedium brevicornu Maxim. and Polygonatum kingianum coll. et Hemsl). METHODS The analysis of Compound Congrong Powder 50% methanolic extract was performed on Agilent HC-C18column( 250 mm × 4. 6 mm,5 μm) maintained at 30 ℃,with acetonitrile( A)- 0. 1% phosphoric acid solution( B) as the mobile phase in a gradient elution manner at a flow rate of 1. 0 m L / min. The detection wavelengths were 330 nm for echinacoside and verbascoside( 0- 20 mim) and 270 nm for icariin( 20- 30 mim). RESULTS The method achieved good linearities in the ranges of 142. 4- 2 848 ng( r = 0. 999 5) for echinacoside,19. 92- 398. 4 ng( r = 0. 999 8) for verbascoside and 19. 60- 392. 0 ng( r = 0. 999 9) for icariin. Their average recoveries( n = 6) were 99. 03%,99. 97% and 99. 50%,respectively. CONCLUSION This method is simple,accurate and reliable,which can be used for the quality control of three components in Compound Congrong Powder.
