摘 要:为研究猪瘟病毒(CSFV)2.1d新基因亚型的基因组特征,本研究根据Gen Bank中登录的Zj0801株CSFV全基因组序列设计合成6对引物,通过RT-PCR方法对2014年分离自山东的两株2.1d亚型CSFV(SDSG1410和SDLS1410)进行全基因组测序,两株CSFV全长均为12 296 nt。全基因组遗传演化分析表明这两个分离株位于一个独立分支中,属于2.1d亚型,与E2基因分型结果一致。核苷酸和推导氨基酸同源性分析显示,分离株全基因组同源性与参考株相比差异较大,与Zj0801株的全基因组同源性最高为97.3%~97.5%,而与1.1亚型的Shimen和HCLV株的同源性仅为85.0%~85.1%和84.3%~84.4%;部分基因和非翻译区变异较大,如N^(pro)、C、E2、NS2、NS5A和3'UTR。对全基因组氨基酸序列分析,结果显示2.1d亚型CSFV在V^(640)、K^(992)、A^(1020)、M^(1090)、R^(1395)和D^(2374) 6处氨基酸上具有共同的分子特征。以上研究结果增加了CSFV 2.1d新基因亚型的分型依据。To investigate the genome characteristic of 2.1d subgenotype of classical swine fever virus (CSFV), two CSFVs named SDSGI410 and SDLS1410 isolated from Shandong area were sequenced with six pairs of primers based on Zj0801 strain of CSFV. Both genomic RNA of the CSFVs were 12,296 nt in length. Phylogenetic analysis indicated that the two CSFVs were classified into 2.1d subgenotype CSFV in comparing with the full viral genomic sequences, which was consistent with the result based on the E2 gene alignment. Then, the nucleotides and the deduced amino acid similarity showed the genome similarity of the two virus, compared with Zj0801, Shimen and HCLV, were 97.3%-97.5%, 85.0%-85.1% and 84.3%-84.4%, respectively. In addition, the high sequence variations were found in N~~, C, E2, NS2, NS5A encoding sequences and 3'UTR regions. Furthermore, the full genome of new 2. ld new subgenotype shared several consistent molecular characteristics such as V640 K992 ,A1020, M1090, R1395 and D2374 in encoding sequence of the virus. These results provided additional evidences other than E2 gene for classification of the new 2. 1d subgenotype of CSFV.
