摘  要:目的:明确T140体内靶向阻断SDF-1/CXCR4信号通路对豚鼠关节软骨Ⅱ型胶原蛋白及聚集蛋白聚糖的影响,探讨T140减缓软骨退变的作用。方法:健康9月龄雄性Hartley豚鼠36只,随机分成A、B、C三组,A组为实验组,B组为实验对照组,C组为空白对照组,每组12只。于12周时,获取膝关节软骨,采用RT-PCR法检测Ⅱ型胶原蛋白(ColⅡ)及聚集蛋白聚糖m RNA的表达;Western blot检测ColⅡ含量。结果:RT-PCR检测结果显示A组Ⅱ型胶原蛋白及聚集蛋白聚糖的m RNA表达量高于B、C组,差异有统计学意义(P<0.05);Western blot检测结果发现A组ColⅡ含量高于B、C组(P<0.05)。结论:T140于体内靶向阻断SDF-1/CXCR4信号通路,减少关节软骨Ⅱ型胶原蛋白及聚集蛋白聚糖的降解,进而减缓关节软骨的退变。Objective The purpose of this study is to explore the effect of T140(an antagonist of SDF-1/CRCR4 signaling pathway)on type Ⅱ collagen and aggrecan in articular cartilage of guinea pigs. Methods 369-month-old male healthy Hartley guinea pigs were divided into three groups:experimental group(A,n=12),experimental control group(B,n=12) and blank control group(C,n=12). Pigs in group A were implanted Alzet capsule type micro osmotic pressure pump containing T140 diluted by PBS solution and in group B implanted the pump containing PBS solution only. Articular cartilages of knee joint were harvested for measuring m RNA levels of type Ⅱ collagen and aggrecan with RT-PCR and protein levels of type Ⅱ collagen with Western blot12 weeks later. Results The m RNA levels of the type Ⅱ collagen and aggrecan in group A were significantly higher than in groups B and C(P〈0.05). The content of type Ⅱ collagen in group A was also significantly higher than in groups B and C(P〈0.05). Conclusion T140 reduces the degradation of type Ⅱ collagen and aggrecan through blocking the SDF-1/CRCR4 signaling pathway and so as decreases the risk of cartilage degeneration.
