牛膝总皂苷对过氧化氢致人脐静脉内皮细胞损伤的保护作用研究

摘  要:目的:观察牛膝总皂苷对过氧化氢(H2O2)诱导人脐静脉内皮细胞(HUVEC)损伤的保护作用。方法:体外培养HUVEC,分为空白对照组、模型对照组及牛膝总皂苷低、中、高浓度组,MTT法检测细胞活力,微板法检测上清液中一氧化氮(NO)的含量,采用ELISA法检测内皮素-1(ET-1)、组织型纤溶酶原激活物(tPA)、纤溶酶原激活物抑制剂(PAI-1)的含量。结果:与模型对照组比较,牛膝总皂苷中、高浓度组细胞活力显著升高,ET-1和PAI-1含量显著减少,NO和tPA含量显著升高(P<0.05或P<0.01)。结论:牛膝总皂苷对H2O2诱导的HUVEC损伤具有显著的保护作用。Objective : To observe the protective effect of Achyranthes bidentata saponins on the injury of human umbilical veinendo- thelial cell (HUVEC) injured by hydrogen peroxide. Methods:The HUVEC were cultured vitro and divided into five groups as fol lows: control group, model group, low-dosage of Achyranthes bidentata saponins group, mid dosage of Achyranthes bidentata sapo- nins capsule group and high-dosage of Achyranthes bidentata saponins capsule group. MTT was used to determine the HUVEC via- bility. The contents of nitric oxide(NO) were measured by the microplate and the method of enzyme-linked immunosorbent assay (ELISA) was used to determin endothelin-1 (ET-1), tissue plasminogen activator (tPA) and plasminogen activator inhibitor-1 (PAI- l). Results: Compared with the model group, the mid-dosage of Achyranthes bidentata saponins and high-dosage of Achyranthes bidentata saponins significantly increased HUVECs viability, decreased the contents of ET-1, PAI 1 and increased the contents of NO and tPA(P〈0.05 or P〈0.01).
