摘 要:目的:研究四逆汤及其不同配伍方对阿霉素诱导大鼠心力衰竭心肌能量代谢的影响。方法:采用腹腔注射阿霉素(2.5 mg/kg)法,每周1次,共6周,建立大鼠慢性心力衰竭模型。将造模成功大鼠随机分为模型组、附子组(1.5 g/kg)、附子-甘草组(3 g/kg)、附子-干姜组(2.5 g/kg)、四逆汤组(4 g/kg)及卡托普利阳性药组(6.25 mg/kg),并设正常对照组。各治疗组连续灌胃给药4周,模型组和正常组予同体积蒸馏水。采用高效液相色谱法测定各组心肌磷酸腺苷(ATP、ADP和AMP)的含量变化。结果:与正常组比较,模型组大鼠ATP含量明显减少(P<0.01),AMP和ADP含量升高(P<0.05);附子-甘草组、附子-干姜组、四逆汤组和卡托普利组较模型组大鼠心肌ATP含量增加(P<0.05),AMP和ADP含量降低(P<0.05)。结论:四逆汤及其不同配伍方可有效调节心衰大鼠的心肌能量代谢紊乱,延缓慢性心衰的发展,以四逆汤全方效应更为明显。Objective: To investigate the effects of Sini Decoction( SND) and its components compatibility on myocardial energy metabolism in rats with chronic heart failure( CHF) induced by doxorubicin. Methods: The CHF rat model was induced by intraperitoneal injection of doxorubicin( ADR,2. 5 mg / kg) once a week for 6 weeks. The surviving model rats were randomly divided into 6 groups: model group,Fuzi group( 1. 5 g / kg),Fuzi Gancao group( 3 g / kg),Fuzi Ganjiang group( 2. 5 g / kg),SND group( 4 g / kg) and captopril group( 6. 25 g / kg). Meanwhile,the normal group was seperately established. All of rats in therapeutic groups were administered the medication orally for 4 weeks and the rats in normal and model groups were given the distilled water with same volume. After treatment for 4 weeks,adenosine triphosphate( ATP),adenosine diphosphate( ADP) and adenosine monophosphate( AMP) in myocardium were examined by high performance liquid chromatography( HPLC). Result: Compared with normal group,the content of ATP in left ventricular muscle was remarked decreased( P〈0. 01) while AMP and ADP were increased in model group( P 〈0. 05). Compared with model group,Fuzi Gancao group,Fuzi Ganjiang group,SND group and captopril group elevated the ATP contents( P 〈0. 05) and decreased AMP and ADP levels( P 〈0. 05). Conclusion: SND and its components compatibility can improve cardiac function and regulate the energy metabolism of heart failure in rats. SND had significant effect on anti-heart failure.
