高效液相色谱-串联质谱法测定益母草颗粒中盐酸水苏碱的含量

摘  要:目的建立高效液相色谱-串联质谱法测定益母草颗粒中盐酸水苏碱含量的方法 。方法 色谱条件采用Agilent SB-C18柱(50 mm×2.1 mm,1.8μm),以乙腈-0.2%冰乙酸(70︰30)为流动相,流速为0.3 ml/min。质谱检测采用正离子电喷雾离子源(ESI+),在多反应监测模式(MRM)下检测,监测离子(m/z)分别为:144/58、144/84。结果盐酸水苏碱浓度在0.2-10.0μg/ml范围内与峰面积呈良好的线性关系(r=0.9992),平均回收率为102.6%,RSD为1.5%(n=6)。结论 该方法快速简便、专属性强、结果准确,可用于益母草颗粒的质量控制。Objective To establish a method of high performance liquid chromatography(HPLC)-tandem mass spectrometry determining the content of stachydrine hydrochloride from Motherwort granule. Methods The chromatographic conditions were Agilent SB-C18column(50 mm×2.1 mm,1.8 μm),acetonitrile to 0.2% glacial acetic acid(70︰30) as the mobile phase,and a flow rate of 0.3 ml/min.Positive ion electrospray ionization(ESI+) was applied for mass spectrometric detection under multiple reaction monitoring(MRM) mode.The monitored ions(m/z) was 144/58,144/84 respectively. Results The concentration of stachydrine hydrochloride being within 0.2 μg/ml to 10.0 μg/ml had a good linear relationship with the peak area.The r value was 0.9992,and the average recovery was 102.6% and the value of RSD was1.5%(n=6). Conclusion The method is fast,simple,specific,and accurate in outcome,which can be used for quality control of Motherwort granule.
